Background

Endothelin-converting Enzyme 1 (ECE-1) is a zinc protease of the neprilysin (NEP) family, which also includes ECE-2, PEX, XCE, DINE, Kell and several NEP-like proteins (1). ECE-1 is a type II transmembrane protein with a short cytoplasmic tail and a large ectodomain. Four alternatively spliced isoforms differ in their cytoplasmic tail (2, 3). In addition to big endothelin-1, ECE-1 cleaves a variety of bioactive peptides such as bradykinin, neurotensin, angiotensin I, and substance P (1). Together with ECE-2, it is also involved in degradation of beta -amyloid peptide (4). The ectodomain of human ECE-1, which is common to all isoforms, was expressed with an N-terminal His tag and purified.

Western Blot: Neprilysin/CD10 Antibody (SN75-07) [NBP2-67717] - Analysis of CD10 on Daudi cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:5,000 dilution was used for 1 hour at room temperature.
Immunocytochemistry/Immunofluorescence: Neprilysin/CD10 Antibody (SN75-07) [NBP2-67717] - Staining CD10 in MCF-7 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

PVDF membrane was probed with 1 µg/mL of Goat Anti-Human DDR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2396) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). Specific bands were detected for DDR1 at approximately 100 and 120 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Simple Western lane view shows lysates of MCF‑7 human breast cancer cell line, loaded at 0.2 mg/mL. A specific band was detected for DDR1 at approximately 137 kDa (as indicated) using 10 µg/mL of Goat Anti-Human DDR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2396) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the12-230 kDa separation system.        
DDR1 was detected in immersion fixed paraffin-embedded sections of human breast cancer tissue using Human DDR1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2396) at 15 µg/mL overnight at 4 °C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue).
