Description
Caspase 4, also known as Ich-2(ICE and CED-3 homolog 2), also known as TX and ICErel-II, can interact with Apaf-1 and participate in caspase 3 activation caused by cytochrome c. Caspase 4 can also interact with caspase 14. This caspase 4 activity assay kit is based on caspase 4 that can catalyze the substrate Ac-LEVD-pNA to generate yellow pNA (p-Nitroaniline). The pNA has a strong absorption near 405 nm, The activity of caspase 4 can be calculated by determining the absorbance at 405 nm.

Background

Caspase 4 encodes a protein that is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes composed of a prodomain and a large and small protease subunit. Activation of caspases requires proteolytic processing at conserved internal aspartic residues to generate a heterodimeric enzyme consisting of the large and small subunits. This caspase is able to cleave and activate its own precursor protein, as well as caspase 1 precursor. When overexpressed, this gene induces cell apoptosis. Alternative splicing results in transcript variants encoding distinct isoforms.
