Western blot shows lysates of human testis tissue. PVDF membrane was probed with 1 µg/mL of Human Autoimmune Regulator/AIRE Antigen Affinity-purified Polyclonal Antibody (Catalog # AF5936) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for Autoimmune Regulator/AIRE at approximately 58 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.

Background

AIRE (Autoimmune Regulator; also APECED) is a 58‑60 kDa member of the PHD Zn-finger family of transcription factors. It appears to have restricted expression, being identified in thymic medullary epithelium, dendritic cells and neutrophils. AIRE regulates the expression of self-antigens in thymus. In particular, genes coding for these antigens are generally inactive, and show minimal H3K4me3 and AcH3 reactivity in their promoters. Human AIRE is 545 amino acids (aa) in length. It contains multiple domains, including an HSR domain that mediates homodimerization (aa 1‑105), an NLS (aa 131‑133), a SAND domain (aa 181‑280), and two Zn-finger PHD domains (aa 296‑343 and 434‑475) that bind to nonmethylated histone H3. There are isoforms that show a Trp substitution for aa 437‑467, a 95 aa substitution for aa 1‑292, and a 75 aa substitution for aa 377‑545. Over aa 476‑545, human AIRE shares 71% aa identity with mouse AIRE.

1. I am looking into a positive control for Western blotting. We are trying to detect the AIRE protein and are not sure whether the antibody we have is picking up the protein. Are you able to tell me whether the human thymus whole tissue lysate (NB820-59456) has been used to detect for AIRE expression by Western blotting?
   • I am not currently aware of any references to the use of our human thymus whole tissue lysate being used to detect AIRE by Western blotting. On looking through the Western blot images for all seven of our AIRE antibodies, I see that these have been generated using lysates from RT-4 cells (bladder), U-251 MG cells (brain), 293 cells (human embryonic kidney), human spleen lysate, liver lysate and tonsil lysate.
