Immunocytochemistry/Immunofluorescence: ADNP Antibody [NBP2-97747] - Immunofluorescence staining of ADNP in U2OS cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-Human ADNP polyclonal antibody (dilution ratio 1:300) at 4C overnight. Then cells were stained with the Alexa Fluor(R)594-conjugated Goat Anti-rabbit IgG Secondary antibody (red). Positive staining was localized to Nucleus.

Immunoprecipitation: ADNP Antibody [NBP2-97747] - ADNP was immunoprecipitated using: Lane A: 0.5 mg U-251MG Whole Cell Lysate. 4 ul anti-ADNP rabbit polyclonal antibody and 60 ug of Immunomagnetic beads. Protein A/G. Primary antibody: Anti-ADNP rabbit polyclonal antibody, at 1:100 dilution. Secondary antibody: Goat Anti-Rabbit IgG (H+L)/HRP at 1/10000 dilution. Developed using the ECL technique. Performed under reducing conditions. Predicted band size: 122 kDa. Observed band size: 150 kDa.

DMS 53 cell line lysates were prepared and immunoprecipitation was performed using 2.0 μg of ADNP Antibody (Catalog # NBP2-97747) pre-coupled to Dynabeads Protein A. Immunoprecipitated ADNP was detected in Western Blot with a Rabbit ADNP antibody used at 1/500. The Ponceau stained transfer of the blot is shown. Western blot shows lysates of DMS 53 parental cell line and ADNP knockdown DMS 53 cell line (KD). Nitrocellulose membrane was probed with ADNP Antibody (Catalog # NBP2-97747) followed by HRP-conjugated secondary antibody. A specific band was detected for ADNP at approximately 135 kDa (as indicated) in the parental DMS 53 cell line, but is significantly reduced in the DMS 53 cell line. Background

The activity-dependent neuroprotective protein (hADNP) gene is frequently amplified in many neoplasias, including breast, bladder, ovarian, pancreatic, and colon cancers. hADNP mRNA is abundantly expressed in distinct normal tissues, and high expression levels were encountered in malignant cells. hADNP is implicated in maintaining cell survival, perhaps through modulation of p53.
