Description
CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

This antibody was raised by immunising LOU rats with soluble recombinant 4-1BB Ligand/TNFSF9:Fc fusion protein.
Specificity
This antibody is specific for 4-1BB ligand (also known as CD137 ligand), which is a member of the TNF superfamily and is a type II membrane protein. 4-1BB ligand is a co-stimulatory molecule that is expressed on antigen presenting cells (DCs, monocytes/macrophages, B cells) and is upregulated upon activation of these cells. Both of the 4-1BB ligand and its receptor, TNFRSF9/4-1BB, are involved in the antigen presentation process and in the generation of cytotoxic T cells. In direct ELISAs, no cross-reactivity with recombinant human RELT is observed.

Background

CD137 exists on the cell surface as a monomer with a molecular mass of 30 kDa and as a dimer of 55 kDa. Human and mouse CD137 share 60% amino acid identity. CD137 (4-1BB), a member of the tumor necrosis factor receptor superfamily, is a type I transmembrane glycoprotein expressed on the cell surface of activated splenic T cells and thymocytes. The functions of CD137 in T lymphocytes include regulating activation, proliferation and apoptosis. CD137 and CD28 are costimulatory molecules of T cell activation. Costimulatory molecules are important in initiating anti-tumor immune responses. CD137 plays an important role in regulating T-cell-dependent immune responses. Expression of CD137 correlates negatively with lymphocyte proliferation and positively with the degree of activation-induced cell death caused by mitogen over stimulation. In monocytes, CD137 induces activation, promotes adherence and prolongs survival.
