Immunoprecipitation (IP) is a common research technique used to precipitate a specific protein (or protein complex) of interest out of a heterogeneous lysate. The concept of precipitating proteins out of solution is often called a "pull-down". There are a variety of methods to achieve immunoprecipitation, the most common of which uses antibodies bound to magnetic or agarose beads. This process can be used to isolate and concentrate a particular protein or complex from a sample containing many thousands of different proteins, and is often used to study protein-protein interactions through Co-IP experiments.

Effects of IGF-1 or/and PDGF-bb on IL-1 beta -induced NF-kappa B-dependent pro-inflammatory, pro-apoptotic and matrix degrading gene products in chondrocytes.To determine whether IGF-1 or/and PDGF-bbexert effects on IL-1 beta -induced NF-kappa B-dependent expression of pro-inflammatory, pro-apoptotic and matrix degrading gene products, primary chondrocytes were either stimulated with 10 ng/ml IL-1 beta , 10 ng/ml PDGF-bb, 10 ng/ml IGF-1 or combination of both growth factors (5 ng/ml each) or pre-stimulated for 12 h with 10 ng/ml PDGF-bb, 10 ng/ml IGF-1 or combination of both growth factors (5 ng/ml each) followed by 10 ng/ml IL-1 beta for 24. Equal amounts of total proteins were separated by SDS-PAGE and analyzed by immunoblotting using antibodies raised against COX-2, MMP-9 and MMP-13 and active caspase-3. Stimulation with IL-1 beta resulted in production of COX-2, MMP-9, MMP-13 and caspase-3 cleavage. Pre-treatment with a combination of both IGF-1 or/and PDGF-bb downregulated COX-2, MMP-9, MMP-13 and cleaved caspase-3. Enhanced MMP13 gene expression and production due to 3D osteogenic differentiation. CTSS attenuates EGF-mediated EGFR degradation.(a) OEC-M1 and MDA-MB-231 cells were pretreated with 20 μM 6r or ZFL for 1 h and subsequently incubated with 100 ng/mL EGF for an additional 2 h. The total cell lysates were analysed using EGFR-specific antibodies. ACTIN was used as the internal control for semiquantitative loading in each lane. (b) The cells were stimulated with EGF (100 ng/mL) with or without the pretreatment of 20 μM 6r for the indicated durations. EGFR degradation was examined through immunostaining by using an anti-EGFR antibody. Notably, a substantial amount of EGFR was detectable even after 6 h of EGF stimulation in 6r-treated cells. (c) The OEC-M1 cells were transiently transfected with plasmids (pCMV) that encoded wild-type CTSS. After 24 h of transfection, the cells were treated with 100 ng/mL EGF for the indicated durations and the cellular EGFR, CTSS, and ACTIN signals were determined through Western blotting. The lifespan of EGF-mediated EGFR degradation was calculated by normalising the signal intensity of EGFR with that of ACTIN. (d) The MDA-MB-231 cells were transfected with specific 50 nM CTSS siRNA (si-CTSS) for 24 h and subsequently incubated with 100 ng/mL EGF for the indicated durations. The nontargeting scramble siRNA (si-SC) was used as the scramble control. (e) The MDA-MB-231 cells were transiently transfected with plasmids encoding the CTSS-C25A mutant. After 24 h of transfection, the cells were incubated with 100 ng/mL of EGF for the indicated durations. Furthermore, the cells were harvested and subjected to SDS-PAGE and Western blotting. EGFR degradation was determined using an antibody against EGFR. ACTIN signalling was included as the loading control. Immunohistochemistry-Paraffin: AG-2/AGR2 Antibody [NBP2-27393] - Analysis using the Azide Free version of NBP2-27393. Staining of human adenocarcinoma of the colon stained with AGR2 antibody (2-5 ug/ml), peroxidase-conjugate and DAB chromogen. Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Western blot shows lysates of mouse kidney tissue. PVDF membrane was probed with 0.25 µg/mL of Goat Anti-Mouse ACE-2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3437) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for ACE-2 at approximately 95 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

HEK293 human embryonic kidney cell line transfected with (A) mouse ACE-2 or (B) irrelevant protein, and eGFP was stained with Goat Anti-Mouse ACE-2 Affinity Purified Polyclonal Antibody (Catalog # AF3437) followed by Allophycocyanin-conjugated Anti-Goat IgG Secondary Antibody (F0108). Quadrant markers were set based on Goat IgG control antibody (AB-108-C, data not shown). Staining was performed using our Staining Membrane-associated Proteins protocol.

A specific band was detected for ACE‑2 at approximately 141 kDa (as indicated) using 2.5 µg/mL of Goat Anti-Mouse ACE‑2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3437) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

Recombinant Mouse ACE‑2 protein was serially diluted 2-fold and captured by Rat Anti-Mouse ACE‑2 Monoclonal Antibody (MAB34371) coated on a Clear Polystyrene Microplate (DY990). Goat Anti-Mouse ACE‑2 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3437) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (DY998) followed by Substrate Solution (DY999) and stopping the enzymatic reaction with Stop Solution (DY994).

The band at 65-70kDa corresponds to ubiquilin 2 protein, which is known to differ between the human and rodent proteins.

Immunocytochemistry/Immunofluorescence: Ubiquilin 2 Antibody (6H9) [NBP2-25164] - Analysis of an NIH-3T3 cell culture stained with mouse mAb to ubiquilin 2, NBP2-25164, dilution 1:1,000 in green, and costained with chicken pAb to lamin A/C, dilution 1:5,000 in red. The blue is DAPI staining of nuclear DNA. The cells were treated with 50uM of chloroquine, an inhibitor of autophagy, for 16 hours prior to staining. NBP2-25164 antibody reveals vesicular staining of ubiquilin 2 protein accumulated in lysosomes in the cytoplasm, while the lamin A/C antibody stains the nuclear lamina.

<b>Genetic Strategies Validation. </b>Immunocytochemistry/ Immunofluorescence: Ubiquilin 2 Antibody (6H9) [NBP2-25164] - HAP1 WT and Ubiquilin 2 KO cells were labeled with a green or a far-red fluorescent dye, respectively. WT and KO cells were mixed and plated to a 1:1 ratio in a 96-well plate with glass bottom. Cells were stained with Ubiquilin-2 antibody (NBP2-25164) and with the corresponding Alexa-fluor 555 coupled secondary antibody including DAPI. Acquisition of the blue (nucleus-DAPI), green (identification of WT cells), red (antibody staining) and far-red (identification of KO cells) channels was performed. $Heat Shock Generates Insoluble Ubiquitin-Positive Aggregates and Does Not Inactive Proteasomes, Related to Figure 1(A) UBQLN2 is not pelleted when cells are heat shocked post lysis. Cell lysates were incubated at 37 or 42C and then fractionated into soluble (S) and pellet (P) fraction. This indicates that UBQLN2 itself does not aggregate as a result of high temperature.(B) UBQLN2 levels are not upregulated in response to heat shock. HSP70 and GAPDH were used as a positive and negative controls, respectively.(C) Heat shock aggregates are insoluble in up to and including 1% SDS but are solubilized in 2% SDS. Blotting of soluble and pellet fractions with anti-ubiquitin and UQBLN2 antibodies confirmed dissolution of the aggregates in 2% SDS.(D) Proteasomes are active after heat shock. To confirm that proteasome activity was not affected by heat shock, we incubated U2OS and MEFs at the indicted temperatures for 2h. Cells were then harvested and cell lysates were incubated with the proteasome inhibitor MG132 or DMSO, followed by incubation with a fluorescent proteasome-activity probe, as indicated. The presence of fluorescently labeled beta-subunits at the same intensity under both heat stress and normal temperature, indicate that proteasome activity is not significantly affected by heat shock.(E) U2OS cells were treated with control or UBQLN2 siRNA and subjected to heat shock for the indicated times. Analysis of the pellet fraction revealed that insoluble ubiquitylated aggregates are generated within 5 min of heat shock, but that depletion of UBQLN2 does not noticeably alter the accumulation of these aggregates at any of the indicated time points.
