Flow Cytometry is a laser based biological assay used for cell detection, counting and sorting of specific biomarkers. Cells are first suspended in solution and fluorescently labeled antibodies are applied. Then the cells are passed single-file through a series of lasers which analyze size, shape and the fluorescent characteristics of each cell. Results can then be electronically gated to filter the data to a specific cell type and whether or not they contain bound antibodies. This form of assay is particularly useful for analyzing blood samples.

$Western Blot: mCherry Antibody (1C51) [NBP1-96752] - Threonine 276 is required for the intramolecular interaction, and for inhibition of membrane binding. Mutation of threonine 276 to aspartic acid promotes the association of cytohesin 2 with membranes. MDCK cells were transfected with constructs encoding mCherry-tagged wild-type or T276D cytohesin 2 and fractionated into cytosol and total membranes. Untransfected HEK293 cells do not express Cherry and do not stain with the antibody, but their nuclei can be visualized using a DNA stain (blue).

Western Blot: mCherry Antibody (1C51) [NBP1-96752] - WB assay of the crude extract of HEK293 cells transfected with pFin-EF1-mCherry vector (lane +) and an equal amount of protein extract from untransfected HEK293 cells (lane -). NBP1-96752 binds a major band running at about 28 kDa (observed molecular weight) corresponding to intact full-length mCherry. The two other bands are clearly processed forms of mCherry as they are not present in non-transfected HEK293 cells.

Immunocytochemistry/Immunofluorescence: GM130/GOLGA2 Antibody [NBP2-53420] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.05% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-GM130/GOLGA2 Antibody NBP2-53420 at 1 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: GM130/GOLGA2 Antibody [NBP2-53420] - HeLa cells were fixed for 10 min using 10% formalin and then permeabilized for 5 min using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-GOLGA2 at 1:200 overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at 1:500. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at 1:500. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: GM130/GOLGA2 Antibody [NBP2-53420] - Total protein from human HeLa and mouse Neuro2A cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 0.5 ug/mL anti-GOLGA2 in 1% non-fat milk in TBST and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Immunohistochemistry-Paraffin: GM130/GOLGA2 Antibody [NBP2-53420] - Analysis of a FFPE tissue section of mouse intesting using GM130/GOLGA2 antibody at 1:1500. The antibody generated staining in the the Golgi apparatus in the perinuclear region.

$Cellular localization of OS-9 & PHD2.(A) A lectin gel-shift assay was conducted to test for glycosylated proteins. Total cell lysates of U2OS cells were incubated in the presence or absence of the endoglycosidases EndoH & PNGaseF for 6 h at 37°C. Digest products were separated on a reducing 10% SDS-PAGE gel which contained concanavalin A co-polymerized in the top layer of the separating gel to retard mobility of glycosylated proteins [56]. Glycosylated OS-9 is indicated as ‘g’, deglycosylated OS-9 as ‘d’. (B) Detection of OS-9 & PHD2 in the nuclear fraction. HEK293 cells with & without transfection of the plasmid pcDNA3-OS-9 were separated into nuclear fraction (N) & postnuclear supernatant (PS), the latter containing cytoplasm & organelles. Western blot analysis included BiP, GAPDH & lamin A as typical marker proteins for the ER, the cytoplasm & the nucleus, respectively. (C) Detection of OS-9 & PHD2 in the cytoplasm. HEK293 cells were co-transfected with pcDNA3-OS-9 & pPHD2-V5. For hypoxia, cells were exposed to 3% O2 for 4 h. To generate nearly anoxic conditions, cells were exposed to an oxygen consuming chemical system or to 1% or 3% O2 for 4 h to generate hypoxia. Western Blot: EGLN1/PHD2 Antibody [NB100-137] - Iron supplementation restores HIF-1 alpha levels to normal following ATP6V0E1 inhibition in HeLa cells. Immunohistochemistry-Paraffin: TNF-alpha Antibody [NBP1-19532] - Analysis of a FFPE tissue section of mouse intestine using TNF-alpha antibody (NBP1-19532) at 1:300 dilution. The binding of this primary antibody to TNF-alpha protein in the section was detected using HRP-labeled secondary antibody and DAB reagent, and nuclei of cells were counterstained using hematoxylin. This TNF-alpha antibody generated an expected diffused immunostaining of this protein in the tested tissue. Staining was primarily observed in the epithelial cells and some cells showed membrane positivity also.

Scutellarin attenuates hypertension-induced brain expression of NF-kappa B, TNF-alpha , IL-1 beta , and IL-18. Western immunoblot analysis for (a) NF-kappa B p65, (b) TNF-alpha , (c) IL-1 beta , and (d) IL-18 in the rat cortex and striatum. Treatment with scutellarin significantly reduced the expression of these inflammatory markers in a dose-dependent manner. *P < 0.001 versus sham group; #P < 0.05, ◆P < 0.001 versus NS group; &P < 0.001 versus low-dose group.

IL-17/IL-17A was detected in immersion fixed paraffin-embedded sections of human tonsil using Goat Anti-Human IL-17/IL-17A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA) at 1 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (VC004). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to lymphocytes. IL-17A mRNA (red) and IL17A protein (green) were detected in formalin-fixed paraffin-embedded tissue sections of human Crohn’s Disease Intestine. ACD’s Integrated Co-Detection Workflow was performed using ACD RNAScope Probe Hs-IL-17A (Catalog # 310931) and goat anti-human IL-17/IL-17A polyclonal antibody (R&D Systems Catalog # AF-317-NA) at 1ug/mL. Tissue was stained using RNAscope® 2.5 HD Detection Kit-RED (Catalog # 322360) and RNAscope® 2.5 LS Green Accessory Pack (Catalog # 322550). Tissue was counterstained with 50% hematoxylin (blue).

<b>Biological Strategies Validation. </b>Human peripheral blood mononuclear cells were unstimulated (light orange filled histogram) or treated with 50 ng/mL PMA and 250 ng/mL Ca 2+ionomycin for 16 hours, then stained with Goat Anti-Human IL-17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA, dark orange filled histogram) or isotype control antibody (AB-108-C, open histogram), followed by Allophycocyanin-conjugated Anti-Goat IgG Secondary Antibody (F0108). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.

Recombinant Human IL-17 (317-ILB) stimulates IL-6 secretion in NIH/3T3 mouse embryonic fibroblasts in a dose-dependent manner (orange line), as measured by the mouse IL-6 Quantikine ELISA Kit (Catalog # M6000B). IL-6 secretion elicited by Recombinant Human IL-17 (15 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human IL-17 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-317-NA). The ND50 is typically 0.02‑0.12 µg/mL.

Expression of psoriasis‐related cytokines in paradoxical psoriasiform reactions. a-Fetoprotein/AFP was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Mouse Anti-Human/Mouse a-Fetoprotein/AFP Monoclonal Antibody (Catalog # MAB1368) at 25 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm.
