Western Blot: BDNF Antibody [NB100-98682] - Human platelet lysate and recombinant mature BDNF. Blocking: 1% LFDM for 30 min at RT; primary antibody (1:1000) incubated at 4C overnight.

Immunohistochemistry-Paraffin: BDNF Antibody [NB100-98682] - Mouse hippocampus. The animal was perfused at a pressure of 130 mmHg with 300 ml 4% FA before being processed for paraffin embedding. HIER: Tris-EDTA, pH 9 for 20 min. Blocking: 0.2% LFDM in TBST filtered thru 0.2 um. Detection was done using Novolink HRP polymer from Leica following manufacturers instructions; DAB chromogen: Candela DAB chromogen. Primary antibody: dilution 1: 1000, incubated 30 min at RT. Sections were counterstained with Harris Hematoxylin.

Western Blot: BDNF Antibody [NB100-98682] - Antagonized TrkB could restrain the activation of astrocytes & microglia & supress the p38/JNK pathway to alleviate the release of IL-1 beta & TNF-alpha in the SDH of CYP-induced cystitis. Western blots showed that the overexpression of a BDNF, b TrkB, cp-TrkB, d Iba1, e GFAP, fp-p38, gp-JNK, h TNF-alpha , & i IL-1 beta were downregulated in comparison to the CYP + DMSO group after ANA-12 treatment. Western Blot: BDNF Antibody [NB100-98682] - BDNF lowered the mechanical withdrawal threshold further & promoted activation of astrocytes & microglia, & enhanced the p38/JNK pathway to aggravate the release of IL-1 beta & TNF-alpha in the SDH of CYP-induced cystitis. a BDNF treated every other day after CYP injection could further lower the mechanical withdrawal threshold & suppress the retrieval of mechanical threshold when compared with the CYP + PBS group. After the exogenous BDNF injection, Western blots showing the expression of b BDNF, c TrkB, dp-TrkB, e Iba1, f GFAP, gp-p38, hp-JNK, i TNF-alpha , & j IL-1 beta were all further upregulated when compared with the CYP + PBS group. Data of mechanical withdrawal threshold were analyzed using a two-way analysis of variance (ANOVA) followed by the Sidak's multiple comparisons test. Compared to that in the control group, the mechanical threshold of the cystitis group decreased significantly after the CYP injection & remained low until day 17, & the minimum threshold value was reached on day 12. Compared to the control group, they were upregulated on days 8, 12, & 17. e Immunofluorescence double staining assay of BDNF & p-TrkB in the SDH. CD34 was detected in immersion fixed paraffin-embedded sections of mouse Thymus using Rat Anti-Mouse CD34, Monoclonal Antibody (Catalog #NB600-1071) at 1:500 dilution at 37 ° Celsius for 2 minutes. Before incubation with the primary antibody, tissue underwent an all-in-one dewaxing and antigen retrieval preprocessing using PreTreatment Module (PT Module) and Dewax and HIER Buffer H (pH 9; Epredia Catalog # TA-999-DHBH).Tissue was stained using the Alexa Fluor™ 647 Goat anti-Rat IgG Secondary Antibody at 1:200 at 37 ° Celsius for 2 minutes. (Yellow; Lunaphore Catalog # DR647RT) and counterstained with DAPI (blue; Lunaphore Catalog # DR100). Specific staining was localized to the membrane. Protocol available in COMET™ Panel Builder.

Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a FFPE tissue section of mouse small intestine using rat anti-mouse CD34 (clone MEC 14.7) at 1:100 dilution. The signal was developed using HRP-conjugated anti-rat secondary with DAB reagent which followed counterstaining of nuclei using hematoxylin. This antibody specifically labelled the endothelial cells in blood vessels located primarily in the sub-mucosa, and of that of the mucosa muscularis and the mucosal lacteal.

Flow Cytometry: CD34 Antibody (MEC 14.7) [NB600-1071] - CD34 (MEC 14.7) antibody was tested at 1:250 in WEHI-3 cells with DyLight 488 (green) alongside a matched isotype control (black).

Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a human renal cancer tissue section using CD34 antibody (clone MEC 14.7) at 1:100 dilution. The antibody did not detect human CD34 which is an expected outcome for this clone. This section was included in NB600-1071's IHC validation testing as a negative control.

Immunohistochemistry-Paraffin: CD34 Antibody (MEC 14.7) [NB600-1071] - Analysis of a FFPE tissue section of human renal cancer xenograft using CD34 antibody clone MEC 14.7 at 1:100 dilution. The signal was developed using HRP-conjugated secondary antibody and DAB reagent followed by counterstaining of nuclei with hematoxylin. Notably, the antibody specifically stained the endothelial cells of the tumor vasculature which is originating from the host animal cells (mouse). This observation was verified by including a negative control in this validation testing (see the IHC image of human renal cancer tissue section with negative signal for CD34).

The antibody specifically generated a staining of the endothelial cells in blood vessels of the colon.

The antibody specifically generated a staining of the endothelial cells in blood vessels of the heart tissue.

Flow Cytometry: CD34 Antibody (MEC 14.7) [NB600-1071] - An intracellular stain was performed on Raw264.7 cells with CD34 Antibody [MEC 14.7] NB600-1071 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rat IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 488 (SA5-10018, Thermo Fisher).

Immunohistochemistry-Paraffin: CCL2/MCP1 Antibody [NBP1-07035] - IHC analysis of a formalin-fixed paraffin-embedded (FFPE) human breast carcinoma tissue section using 1:1000 dilution of CCL2/MCP1 antibody (NBP1-07035) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) with 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching using peroxide block. The sections were incubated with primary antibody for 30 minutes. Bond Polymer Refine Detection (Leica Biosystems) and DAB were used for signal detection which followed counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) were performed using Aperio AT2 (Leica Biosystems). This antibody generated a diffused cytoplasmic staining of CCL2 antigen in the cancer cells, stromal cells as well as the endothelial cells. The stroma itself showed a weak immunopositivity for CCL2. Staining was performed by Histowiz.

Ability of the antibodies to capture CD44 antigen was first assessed by comparing the level of CD44 antigen from the starting material (SM) to its level remaining in the unbound fractions (UB). Immunohistochemistry-Paraffin: HIF-1 alpha Antibody [NB100-654] - IHC analysis of a formalin-fixed paraffin-embedded tissue section of human endometrium carcinoma AN3CA cell line based xenograft using rabbit polyclonal HIF-1 alpha antibody NB100-654 at 1:300 dilution. The signal was developed using HRP-labelled secondary antibody and DAB reagent, and the section was further counterstained using hematoxylin. The tested section depicted mainly a diffused cytoplasmic staining but there were some cells which showed nuclear signal also (representing hypoxic cells).

Simple Western: HIF-1 alpha Antibody [NB100-654] - Simple Western lane view shows a specific band for HIF-1 alpha in 0.5 mg/ml of Hypoxic HeLa lysate. This experiment was performed under reducing conditions using the 12-230 kDa separation system.

Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). Supershift assay showed that the HIF-1/DNA complex was shifted up in the presence of wild-type (wt) oligo probe and 4 ug HIF-1 alpha antiboby (lanes #13 & 14). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature.
