Application Notes
This YB1 polyclonal antibody is useful for Immunocytochemistry/Immunofluorescence, Western blot, Immunohistochemistry on paraffin embedded sections and Immunoprecipitation. In ICC/IF, cytoplasmic staining was observed in HeLa cells. In WB, a band is seen at approx. 50 kDa representing YB1. In IHC-P, staining was observed in the cytoplasm, nucleus, and nucleoli of mouse testis, brain and kidney tissues. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

In Simple Western only 10 - 15 uL of the recommended dilution is used per data point. Separated by Size-Wes, Sally Sue/Peggy Sue.
The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. Use in vivo reported in scientific literature (PMID: 11447123).

Background

YB1, also known as YBX1 (Y box binding protein 1) or DbpB (DNA binding protein B), is a transcription factor that binds single-stranded DNA within the reverse CCAAT promoter element. YB1 is found abundantly in the cytoplasm and nucleus of many cell types. It binds to cytoplasmic mRNA and participates in translation regulation. Additionally, it may play a role in DNA repair. Current research has shown that YB1 is overexpressed in many different types of cancerous tissues including non-small cell lung carcinoma, breast cancer, colorectal cancer, and ovarian adenocarcinomas.
