YB1 Antibody

Knockdown Validated: YB1 Antibody [NBP1-97572] - Western blot shows lysates of HeLa human cervical epithelial carcinoma parental cell line and YB-1 knockout (KO) HeLa cell line. PVDF membrane was probed with 1:10000 ug/ml of Rabbit Anti-Human YB-1 Polyclonal Antibody (Catalog # NBP1-97572) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for YB-1 at approximately 49 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

Western Blot: YB1 Antibody [NBP1-97572] - Cytoplasmic and nuclear distribution of Ataxin-2, G3BP1, HuR, TIA-1, TIAR, and YB1 (NBP1-97572). Huh7 cells were mock infected or infected with the Cambodia strain of ZIKV at an MOI of 5, and 1 day postinfection the cytoplasmic and nuclear fractions were isolated. The distribution of ZIKV NS5, Ataxin-2, G3BP1, HuR, TIA-1, TIAR, YB1, GRP78, and Fibrillarin was analyzed by Western blotting. Immunocytochemistry/Immunofluorescence: YB1 Antibody [NBP1-97572] - The YB1 antibody was tested in HeLa cells with Dylight 488 (green). Nuclei and alpha-tubulin (NB100-690) were counterstained with DAPI (blue) and Dylight 550 (red).

• Application Notes: This YB1 polyclonal antibody is useful for Immunocytochemistry/Immunofluorescence, Western blot, Immunohistochemistry on paraffin embedded sections and Immunoprecipitation. In ICC/IF, cytoplasmic staining was observed in HeLa cells. In WB, a band is seen at approx. 50 kDa representing YB1. In IHC-P, staining was observed in the cytoplasm, nucleus, and nucleoli of mouse testis, brain and kidney tissues. Prior to immunostaining paraffin tissues, antigen retrieval with sodium citrate buffer (pH 6.0) is recommended.

Background

YB1, also known as YBX1 (Y box binding protein 1) or DbpB (DNA binding protein B), is a transcription factor that binds single-stranded DNA within the reverse CCAAT promoter element. YB1 is found abundantly in the cytoplasm and nucleus of many cell types. It binds to cytoplasmic mRNA and participates in translation regulation. Additionally, it may play a role in DNA repair. Current research has shown that YB1 is overexpressed in many different types of cancerous tissues including non-small cell lung carcinoma, breast cancer, colorectal cancer, and ovarian adenocarcinomas.
