1. I have tested this antibody and would like to share the results from my first preliminary experiment. This Ab has been tested in ICC using a human cell line. Blocking buffer contains either goat or donkey serum. Dilutions: both 1:1000 and 1:2000 have been tested. Secondary ab is donkey anti mouse Alexa-488 (1:900). Results: Hardly any signals to be detect. As I failed to detect any signals, I will increase the concentration by using the following dilutions instead: 1: 200 and 1:500. If I still fail to detect TRPC4, in addition to the absence of the primary antibody, any else I need to do in order to confidently to say that the TRPC4 protein is NOT expressed in the human cell line examined?
   • I don't know what your samples are, but I would check with the literature to determine if it is expected to be expressed. Some preliminary information can be found on UniProt (https://www.uniprot.org/uniprotkb/Q9UBN4/entry). To determine if the sample is truly negative, I would recommend confirming the negative antibody staining with PCR. You should also consult with your mentor regarding these results as they will have more knowledge about your experiments than I will.
