Immunocytochemistry/Immunofluorescence: Skp1 Antibody (JM27-37) [NBP2-67147] - Staining Skp1 in SH-SY5Y cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: Skp1 Antibody (JM27-37) [NBP2-67147] - Immunohistochemical analysis of paraffin-embedded human breast tissue using anti-Skp1 antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: Skp1 Antibody (JM27-37) [NBP2-67147] - Analysis of Hela cells with Skp1 antibody at 1/100 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black).

Background

Skp1 encodes a component of SCF complexes, which are composed of this protein, cullin 1, a ring-box protein, and one member of the F-box family of proteins. This protein binds directly to the F-box motif found in F-box proteins. SCF complexes are involved in the regulated ubiquitination of specific protein substrates, which targets them for degradation by the proteosome. Specific F-box proteins recognize different target protein(s), and many specific SCF substrates have been identified including regulators of cell cycle progression and development. Studies have also characterized the protein as an RNA polymerase II elongation factor. Alternative splicing of this gene results in two transcript variants. A related pseudogene has been identified on chromosome 7.
