Background

C1 Inactivator is a plasma protein that plays a pivotal role in the regulation of the early stages of the classical complement pathway and the contact activation pathways. It is a protease inhibitor acting as a competitive substrate for its target enzymes. These cleave a 34 amino acid peptide, the remainder of the C1 inactivator molecule binding to the protease causing irreversible inhibition. C1 inactivator is one of only two known enzymic inhibitors of C1s, the other, nexin I, is far less effective. The mean serum concentration of C1 inactivator is 0.2g/L. C1 inactivator consists of a single heavily-glycosylated polypeptide chain with a molecular weight of 104 kDa.
