Background

Scavenger receptor class A member 5 (SCARA5), also known as TESR, is part of the scavenger receptor superfamily, which is composed of many members with diverse structures, expression patterns, and functions. SCARA5 is a cell-surface receptor that contains that collagen-like domain and N-terminal scavenger receptor cysteine rich domain typical of the class A scavenger receptor subfamily. Unlike other class A receptors, SCARA5 does not endocytose acetylated or oxidized low density lipoprotein. Instead, SCARA5 binds serum ferritin and mediates its endocytosis from the cell surface, indicating SCARA5 may play a role in non-transferrin iron delivery. At least four isoforms of SCARB1 are known to exist.

  NOTE:- Dr Zhong (corresponding author) confirmed Novus Biologicals that the catalog number of SCARA5 antibody used in this publication is NBP1-77050. This SCARA5 antibody was used for IHC-P staining of paired samples of oral squamous cell carcinoma tissues (collected at Ninth People’s Hospital, Shanghai Jiao Tong University School of Medicine). Sections were deparaffinized through heating the slides at 65 C for 2 hours and treatment with xylene, and the tissues were rehydrated with decreasing concentrations of ethanol. Rabbit polyclonal SCARA5 antibody was used at 1:500 dilution with 4C overnight incubation and then was visualized with DAB detection method via goat anti-rabbit secondary antibody. Positive expression grade and percentage scores were considered for evaluating SCARA5. The authors also used this SCARA5 antibody for WB analysis of 20-40ug lysates from clinical samples as well as from SCARA5 siRNA transfected HN13, HN30, and HIOEC cell lines.

1. We are trying to detect chicken SCARA5 protein by western blot using NBP1-77050 antibody (Antibody was raised against a 17 amino acid peptide near the carboxy terminus of human SCARA5). I am wondering whether the antibody can detect the chicken SCARA5 protein based on homology between human and chicken. Can I get AA sequences of the antigenic peptide?
   • I have now heard back from the lab in response to your enquiry. Although they cannot disclose the amino acid sequence of the antigenic peptide, they have analyzed how the immunogen compares to the equivalent sequence in chicken, and there is 70% homology. Unfortunately, based on this percentage and distribution of homologous amino acids, it is difficult to predict whether the antibody will be able to detect the chicken SCARA5 protein.
