Background

TINAGL1 (Tubulointerstitial nephritis antigen-like 1; also TIN-ag-RP, AZ-1 and Lipocalin 7) is a 54-57 kDa glycoprotein member of the peptidase C1 family of molecules. Mature human TINAGL1 contains an SMB (somatomedin B) domain, one vWFC domain, and a nonenzymatic peptidase C1A region (1). Mature human TINAGL1 shares 90% amino acid (aa) identity with mouse TINAGL1. Alternative splicing generates additional isoforms that either lack a portion of the vWFC domain or consist of a portion of the peptidase domain with a 38 aa insertion and flanked by substitutions (2). TINAGL1 is secreted by renal tubule epithelium, zona glomerulosa cells, vascular smooth muscle, and striated muscle cells (1, 3, 4). It is also expressed in mouse decidual endometrium and blastocysts (5-7). During pregnancy it binds to Laminin-1 and Integrins alpha 5 and beta 1 and plays a role in supporting fertility (6-8). In the vasculature, TINAGL1 localizes to basal laminae along adrenocortical sinusoidal capillaries where it binds to fibronectin and collagen and supports adrenocortical cell adhesion (9).
