Notes

HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.This product is manufactured by and sold under license from OriGene Technologies and its use is limited solely for research purposes.

Background

Alzheimer's disease (AD) patients with an inherited form of the disease carry mutations in the presenilin proteins (PSEN1; PSEN2) or in the amyloid precursor protein (APP). These disease-linked mutations result in increased production of the longer form of amyloid-beta (main component of amyloid deposits found in AD brains). Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Several alternatively spliced transcript variants encoding different isoforms have been identified for this gene, the full-length nature of only some have been determined. [provided by RefSeq].   1. I purchased your product Presenilin 1 Lysate (NBL1-14860) this year. I would like to know if you could tell me the exact concentration of Presenilin-1 present in such lysate, since the concentration given (1mg/ml) is for the total protein content. It would be really helpful for our experiments ongoing at the moment regarding AD.
    • The Presenilin 1 lysate was produced in HEK293T cells using plasmid ID RC216443. The concentration of the lysate protein is 1mg/ml however, I'm unable to confirm the exact amount or concentration of the overexpressed protein. The protein contains a C-terminal DDK tag and its presence in the lysate was confirmed by Western blot.
