Application Notes
Western blot: Preliminary experiments gave an approx 75Da band in Human Heart lysates after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 58.5kDa according to NP_006203.2 and 54.4kDa according to NP_001018063.1. The 75kDa band was successfully blocked by incubation with the immunizing peptide

ECM‑1 was detected in immersion fixed paraffin-embedded sections of human colon cancer tissue using 15 µg/mL Sheep Anti-Human ECM‑1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3937) overnight at 4 °C. Tissue was stained with the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Specific labeling was localized to the plasma membrane of epithelial cells. Lower panel shows a lack of labeling if primary antibodies are omitted and tissue is stained only with secondary antibody followed by incubation with detection reagents. Flow Cytometry: AMPK alpha 1 Antibody (2B7) [NBP2-22127] - An intracellular stain was performed on RH-30 cells with AMPK alpha 1 Antibody (2B7) NBP2-22127PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Phycoerythrin.
