HEK293T cells in 10-cm dishes were transiently transfected with a non-lipid polymer transfection reagent specially designed and manufactured for large volume DNA transfection. Transfected cells were cultured for 48hrs before collection. The cells were lysed in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix, 1mM PMSF and 1mM Na3VO4, and then centrifuged to clarify the lysate. Protein concentration was measured by BCA protein assay kit.This product is manufactured by and sold under license from OriGene Technologies and its use is limited solely for research purposes.

Background
Voltage-gated proton (hydrogen) channels play an important role in cellular defense against acidic stress. They are unique among ion channels with respect to their extremely high selectivity, marked temperature dependence, and unitary conductance, which is 3 orders of magnitude lower than that of most other ion channels. NOX1 is a homolog of the catalytic subunit of the superoxide-generating NADPH oxidase of phagocytes, gp91phox. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]. Transcript Variant: This variant (NOH-1L) represents the longer transcript and encodes the longer protein (isoform long). My question is about in what buffer and which volume I can dissolve the lysate. The data sheet remarks that the concentration is 1 mg/ml.
- This should be shipped as a solution of 100ug (100ul) at 1mg/ml total protein in modified RIPA buffer (25mM Tris-HCl pH7.6, 150mM NaCl, 1% NP-40, 1mM EDTA, 1xProteinase inhibitor cocktail mix (Sigma), 1mM PMSF and 1mM Na3VO4. The lysate is shipped with dry ice. Upon receiving, store the sample at -20C. Lysate samples are stable for 12 months from date of receipt when stored at -20C. Avoid repeated freeze-thaw cycles. Lysate samples can be diluted with 2xSDS Sample Buffer. After dilution, the protein sample should be aliquoted and stored at -20C for long term storage. Prior to SDS-PAGE fractionation, boil the lysate for 5 minutes. We recommended loading 5ul of lysate mixed with 5ul of 2xSDS Sample buffer per lane of each gel for initial testing.
