After speaking with your Tech Support, I tried the antibody (NBP1-81002) for western blotting, which is not one of the applications listed. The protein is Nicalin. Loading 10 ug of mouse brain lysate, I received a distinct band at the expected molecular weight (~ 63 kDa), very clean. Taking note of the Nicalin antibody you offer that can indeed be used for western blotting (NBP2-19493), the spec sheet recommended 1:500-1:3000. Also taking note of the Nicalin antibody that I was trying (NBP1-81002), the concentration being 0.05 ug/ul, I used an equivalent quantity of antibody to test it at the 1:500 dilution (this required I use 40 ul of antibody in 1 ml blocking buffer). While use of such might make this antibody not economical, it does indeed work for western blotting. Please let me know what other information you need to post a review. The gel was 7.5%, the primary antibody was incubated O/N at 4 degrees in 5% milk buffer/0.05% Tween 20; the marker is Magic Mark XP from Invitrogen, the bands being, from HMW to LMW, in kDa as follows: 220, 120, 100, 80, 60, 50, 40, 30 (with 20 immediately below). The secondary antibody was a donkey anti-rabbit HRP-conjugate used at 1:7,500. The exposure is 20 min.
