Background

Neurogenin 1 is the protein product of an activator-type bHLH gene (Ngn1). The activator-type bHLH genes (which also include Mash1 and Math) promote neuronal fate determination and induce expression of Notch ligands such as Delta which activate Notch signaling. The activator type bHLH genes are antagonized by repressor-type basic helix-loop-helix (bHLH) genes such as Hes1, 3 and 5. Thus, activator-type and repressor-type bHLH genes regulate each other, allowing only subsets of neural stem cells to undergo differentiation. Neurogenin 1 is thus one of the key regulators of neural stem cell differentiation, which is essential for generation of complex brain structures of appropriate size, shape and cell arrangement.
