• Application Notes: WB: Preliminary experiments gave bands at approx. 60 kDa band and 35 kDa band in human testis, ovary and uterus lysates and in mouse and rat testis lysates after 0.1 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 105 kDa band according to NP_002431.2. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).

Background

The Escherichia coli MutHLS system has been highly conserved throughout evolution. The eukaryotic pathway results in a specialization of MutS homologs such as MSH2 that have evolved to play crucial roles in both DNA mismatch repair and meiotic recombination. In Saccharomyces cerevisiae, MSH4 is a meiosis-specific protein that is not involved in mismatch correction. This protein is required for reciprocal recombination and proper segregation of homologous chromosomes at meiosis I.
