Immunohistochemistry-Paraffin: SCP3/SYCP3 Antibody [NB300-232] - IHC analysis of a formalin fixed and paraffin embedded tissue section of Mouse testis using SCP3 antibody at 1:1000 dilution. The signal was detected using HRP-DAB detection method and the nuclei were counterstained with hematoxylin. The antibody generated a specific signal of SCP3 in the spermatogonial cells and the spermatocytes. The signal was strongest in the nuclei of spermatogonial cells.

Background

SCP3 (Synaptonemal complex protein 3) is a DNA-binding nuclear protein that belongs to the XLR/SYCP3 family and is an essential structural component of the transverse filaments of synaptonemal complexes formed between homologous chromosomes during meiotic prophase. The synaptonemal complex develops during prophase of first meiosis, wherein it allows the synapse as well as recombination between maternal - paternal chromosomes, and SCP3 is essential to hold cohesin complex between homologous chromosomes, which avoid premature segregation thereby preventing aneuploidies. SCP3 is involved in phosphorylation of HORMAD1 and HORMAD2. It is essential to development of testis (where it is primarily localized) and defective SCP3 may leads to spermatogenic failure type 4 (SPGF4). SCP3 deificient mouse depicts high numbers of aneuploidies in oocytes and offspring. SCP3 is also expressed in certain malignancies including human acute lymphatic leukemias, cervical cancer cells, lung cancer cells etc.

  SCP3/SYCP3 antibody was used for IHC on cryosections of paired female gonads at HH38 and HH42 from fertilized White Leghorn chicken (Gallus gallus) eggs, and the assay implicated fixation of sections for 10?minutes at room temperature in a 4% PFA-PBS solution, ON cryoprotection at 4°C in 30% sucrose in PBS, 10um OCT sections, blocking with1% bovine serum albumin, use of primary antibody at 1:500 dilution (See full text for more details and the staining images may be found in Figure 3, Figure 4, and Figure S2/S4).

• Special Applications: The antibody works in adult marmoset testis but not in neonate nor juvenile testis as spermatogensis has not taken place. However, we only observe the expression in the spermatozoa, which causes a bit concern. This is because that SCP3 was reported to be expressed in the spermatocyte, which was not observed in adult marmoset testis.
