MRP1 Antibody (MRPm5) Summary

Immunogen
The antibody reacts with the MRP1 antigen.
Specificity
MRPm5 reacts with an internal epitope of MRP1, a 180-195 kD transmembrane transporter protein overexpressed in various human non-P-glycoprotein MDR tumor cell lines. MRPm5 was raised against a bacterial fusion protein of MRP1, containing amino acids 986-1204 of the protein. MRPm5 does not cross-react with the human MDR1 and MDR3 gene products.

Application Notes
MRPm5 has potential value for detection of MRP MDR in human tumor samples. Immunocytochemistry: use 1:20-50 dil. on acetone or paraformaldehyde (4% in PBS) fixed cytospin preparations. For immunohistochemistry: MRPm5 (use 1:20) on acetone fixed frozen sections can be followed by incubation with rabbit anti-mouse IgG (1:25) and a monoclonal mouse APAAP complex (1:50). MRPm5 can also be used on formaldehyde-fixed paraffin-embedded human tissues and tumours, after pretreatment with 0.01 M citric acid (pH 6.0) in distilled water at 100 C for 10 min. Flow cytometry: use 1:20-50 dil. after fixing cells in 10% Lysing solution in H2O, and anti-mouse-FITC. Western blotting: use 1:20-50 dil, and anti-mouse-HRP.

Background

The MRP molecule, an integral membrane glycophosphoprotein belonging to the same superfamily of ATP-binding cassette transmembrane transporter proteins as P-glycoprotein, is overexpressed in many P-glycoprotein-negative, multidrug resistant cell lines and tumours. It is believed that the main function of MRP in drug resistance is that of a plasma membrane drug efflux pump.
