| Application Notes | MRPm5 has potential value for detection of MRP MDR in human tumor samples. Immunocytochemistry: use 1:20-50 dil. on acetone or paraformaldehyde (4% in PBS) fixed cytospin preparations. For immunohistochemistry: MRPm5 (use 1:20) on acetone fixed frozen sections can be followed by incubation with rabbit anti-mouse IgG (1:25) and a monoclonal mouse APAAP complex (1:50). MRPm5 can also be used on formaldehyde-fixed paraffin-embedded human tissues and tumours, after pretreatment with 0.01 M citric acid (pH 6.0) in distilled water at 100 C for 10 min. Flow cytometry: use 1:20-50 dil. after fixing cells in 10% Lysing solution in H2O, and anti-mouse-FITC. Western blotting: use 1:20-50 dil, and anti-mouse-HRP. |
