Background

Myristolyated alanine rich C-kinase substrate, hence MARCKS, was originally discovered as a major substrate for protein kinase C in the brain and other tissues, and was originally isolated from human epithelial cells. MARCKS is a major protein of brain, and is concentrated in synapses of neurons. It appears to function in synaptic vesicle cycling and has been shown to bind both actin and calmodulin in vitro. MARCKS belongs to a family of proteins with similar actin and calmodulin binding properties. Deletion of the MARCKS gene in mice results in embryonic brain defects and death.

Western blot shows lysates of mouse brain (cortex) tissue, human brain (motor cortex) tissue, and rat brain (cortex) tissue. PVDF membrane was probed with 0.5 µg/mL of Sheep Anti-Human/Mouse/Rat Neurogranin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7947) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Neurogranin at approximately 14-16 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Neurogranin was detected in perfusion fixed frozen sections of rat brain (hippocampus) using Sheep Anti-Human/Mouse/Rat Neurogranin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7947) at 0.5 µg/mL overnight at 4 °C. Tissue was stained using the NorthernLights™ 557-conjugated Anti-Sheep IgG Secondary Antibody (red; Catalog # NL010) and counterstained with DAPI (blue). Recombinant Human Neurogranin protein was serially diluted 2-fold and captured by Mouse Anti-Human Neurogranin Monoclonal Antibody (Catalog # MAB79473) coated on a Clear Polystyrene Microplate (Catalog # DY990). Sheep Anti-Human/Mouse/Rat Neurogranin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7947) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (Catalog # DY998) followed by Substrate Solution (Catalog # DY999) and stopping the enzymatic reaction with Stop Solution (Catalog # DY994).
