Immunohistochemistry-Paraffin Intended Use: This product is intended for qualitative immunohistochemistry with normal and neoplastic formalin-fixed, paraffin-embedded tissue sections, to be viewed by light microscopy. Clinical interpretation of staining results should be accompanied by histological studies with proper controls. Patients? clinical histories and other relevant diagnostic tests should be utilized by a qualified person (s) when evaluating and interpreting results. Staining Protocol: We suggest an incubation period of 30 minutes at room temperature. Optimal incubation conditions should be determined by the user based upon the fixation conditions and staining system employed. Suitable for formalin fixed, paraffin embedded tissue sections. |

Background
Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. Its protein product is a protease present in seminal plasma. It is thought to function normally in the liquefaction of seminal coagulum, presumably by hydrolysis of the high molecular mass seminal vesicle protein. Serum level of this protein, called PSA in the clinical setting, is useful in the diagnosis and monitoring of prostatic carcinoma. Alternate splicing of this gene generates two transcript variants encoding different isoforms. Additional transcript variants have been described, but it is unclear if these transcripts are normally expressed or if they are specific to benign or malignant tumors. Its expression is apparently confined to the duct cells of the prostate and as such is potentially useful as a serum marker for the treatment of prostate derived tumours. It is a member of the kallikrein subgroup of the (chymo)trypsin serine protease family, but differs from the prototypical member of this subgroup, tissue kallikrein, in possessing a specificity more similar to that of chymotrypsin than trypsin. The gene is located 12kb upstream from the hGK-l gene on chromosome 19. It hydrolyzes semenogelin-1 thus leading to the liquefaction of the seminal coagulum. Alternate splicing of this gene generates several transcript variants encoding different isoforms.
