Background

IRS2 is a cytoplasmic signaling molecule that mediates effects of insulin, insulin-like growth factor 1 (IGF1), and other cytokines by acting as a molecular adaptor between diverse receptor tyrosine kinases and downstream effectors. IRS2 is phosphorylated by the insulin receptor tyrosine kinase upon receptor stimulation, as well as by an interleukin 4 receptor-associated kinase in response to IL4 treatment. IRS2-deficient mice display all the characteristics of type 2 diabetes, suggesting that dysfunction of the IRS2 gene may contribute to the pathogenesis of human type 2 diabetes. Recent work has demonstrated a critical role for IRS2 in the maintenance of peripheral insulin sensitivity, central leptin sensitivity, proper beta cell development in the islets of Langerhans and expansion of functional beta-cell mass in vivo.
