WB: Approx. 120 kDa band observed in A431, Daudi and HeLa lysates (calculated MW of 120 kDa band according to NP_006382.1). An additional band of unknown identity was also consistently observed at 26-28 kDa. This band was successfully blocked by incubation with the immunizing peptide.

The importin-alpha/beta complex and the GTPase Ran mediate nuclear import of proteins with a classical nuclear localization signal. The protein encoded by this gene is a member of a class of approximately 20 potential Ran targets that share a sequence motif related to the Ran binding site of importin beta. Similar to importin beta, this protein prevents the activation of Ran's GTPase by RanGAP1 and inhibits nucleotide exchange on RanGTP, and also binds directly to nuclear pore complexes where it competes for binding sites with importin beta and transportin. This protein has a Ran-dependent transport cycle and it can cross the nuclear envelope rapidly and in both directions. At least four importin beta like transport receptors, namely importin beta itself, transportin, RanBP5 and importin 7, directly bind and import ribosomal proteins.

C4-2 human prostate cancer cell line,LNCaP human prostate cancer cell line
Four siRNAs designed to target the ORF of Importin 7 were transfected into C4-2 human prostate cancer cells (~80% confluence) in a 6-well plate, along with non-target siRNA and transfection reagent-only controls. Cells were lysed with RIPA buffer after 3 days of incubation followed by SDS-PAGE on 10% polyacrylamide gel and transfer to PVDF membrane. Membrane was blocked with 5% milk for 1 hr, washed with 1X TBST, and incubated with 1:1000 dilution of anti-importin 7 primary antibody (NB100-1081) in 3% BSA overnight at 4 degrees Celsius. Membranes were washed with 1X TBST, and incubated with 1:500 dilution of mouse anti-goat secondary antibody in 5% milk for 1 hr at room temperature. Loading control was measured by incubating with 1:10,000 anti-GAPDH-HRP in 1X TBST for 30min.

I have noticed that you have a rabbit polyclonal anti-human Ipo7. I am searching for a primary mouse/goat ab anti-human Importin7 (Ipo7) that is suitable for IF, WB application. Could you please tell me if you have something that is suitable for me?
NBP1-87880 suits all of your needs but is a rabbit host. I am not sure if this will work for you or not. NB100-1081 is our only goat host antibody and it has been validated in Western blot and ELISA. It is expected to work in ICC though, since it is known to recognize the protein in its native form. Please let me know if you have further questions or need further clarification.
