Background

Heat shock protein 90-1a (HSP90-1a) is a myosin chaperone involved in heat shock responses, as well as in myofibrillogenesis. It is known to interact with histone methyltransferases, influencing epigenetic changes in certain pathways. It interacts with TERT to assemble the telomerase holoenzyme complex, which is critical for addition of DNA to telomeric ends. It also is important in thick and thin filament organization, influencing the formation and maintenance of sarcomere characteristics. It is highly expressed in skeletal and cardiac muscles.

Western Blot: GLS2 Antibody [NBP1-76544] - Glutaminase protein expression in PC3 and PC3M cell lines before/after CB-839 treatment. PC3 & PC3M cells were treated (+) or not treated (-) with GLS1 inhibitor CB-839 (1 uM) and their expression of GLS1 & GLS2 proteins determined by Western blot. Antibodies specific for GAC & KCA were utilized on two replicate samples, while GLS2 expression was determined on three. PC3 cell lysates expressed GLS2 more than PC3M cell lysates regardless of CB-839 treatment. The difference in GLS2 expression was significant in the untreated cells (P?<?0.03) but because of high variability the difference was not significant in the drug-treated cells. Levels of GLS1 (GAC and KGA) were similar in all cell lysates. Immunohistochemistry-Paraffin: RRAS Antibody - Unpurified [NB100-56161] - RRAS was detected in immersion fixed paraffin-embedded sections of human placenta using this antibody at 1:500 for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC003). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to the cytoplasm and plasma membrane in decidual and trophoblast cells.

Knockdown Validated: Tmp21/p23 Antibody [NB110-57586] - Western blot shows lysates of HEK293T human embryonic kidney parental cell line and TMP21/TMED10 knockout (KO) HEK293T cell line. PVDF membrane was probed with 2.0 ug/ml of Rabbit Anti-Human TMP21/TMED10 Polyclonal Antibody (Catalog # NB110-57586) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for TMP21/TMED10 at approximately 25 kDa (as indicated) in the parental HEK293T cell line, but is not detectable in the knockout HEK293T cell line. This experiment was conducted under reducing conditions.

Glucose-Regulated Protein 94 (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with an antibody recognizing Glucose-Regulated Protein 94 at a dilution of 1:100 over night at 4C, washed with PBS and incubated with a DyLight-488 conjugated.
