Human GRO2 Luciferase - (LUCPorter™) Stable Reporter Cell Line Summary
Description |
The CXCL2 Prom/LUCPorter™ reporter cell line can be used for screening of agonists, antagonists or signaling inhibitors related with the CXCL2 induction-related signaling pathways. CXC-motif chemokine ligand 2 (CXCL2) is a small cytokine that belongs to the C-X-C chemokine family and is also known as macrophage inflammatory protein 2 (MIP-2). CXCL2 is one of the major proinflammatory cytokines which is induced by innate immune receptors such TLRs and NLRs and also mediates LPS-induced osteoclastogenesis. The CXCL2 induction by phorbol 12-myristate 13-acetate (PMA) as well as flagellin is shown in Figures 1 and 2. In the test, PMA and flagellin induced the CXCL2 promoter activity in a dose response manner, which was quantified by luciferase activity (Figures 1 and 2).

Notes
Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed.
- Use pipette aids to prevent ingestion and keep aerosols down to a minimum.
- No eating, drinking or smoking while handling the CXCL2 reporter cell line.
- Wash hands after handling the CXCL2 reporter cell line and before leaving the lab.
- Decontaminate work surface with disinfectant or 70% ethanol before and after working with the cell line.
- All waste should be considered hazardous.
- Dispose of all liquid waste after each experiment and treat with bleach.
