Background

Gabpb2 encodes the GA-binding protein transcription factor, beta subunit. This protein forms a tetrameric complex with the alpha subunit, and stimulates transcription of target genes. The encoded protein may be involved in activation of cytochrome oxidase expression and nuclear control of mitochondrial function. The crystal structure of a similar protein in mouse has been resolved as a ternary protein complex. Multiple transcript variants encoding distinct isoforms have been identified for this gene.

Western blot shows lysates of Jurkat human acute T cell leukemia cell line, human thymus tissue, mouse thymus tissue, and rat CD4+ T cells. PVDF membrane was probed with 0.5 µg/mL of Sheep Anti-Human/Mouse/Rat Ets-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7284) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Ets-1 at approximately 54 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Ets‑1 was detected in immersion fixed paraffin-embedded sections of human ovary using Sheep Anti-Human/Mouse/Rat Ets‑1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF7284) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Sheep IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC006). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to plasma membrane in epithelial cells. Immunocytochemistry/Immunofluorescence: ERG Antibody [NBP2-60655] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with anti-ERG at 5 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
Western Blot: ERG Antibody [NBP2-60655] - Total protein from Human THP-1, Jurkat and K562 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-ERG in blocking buffer and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.
Immunohistochemistry-Paraffin: ERG Antibody [NBP2-60655] - IHC analysis of a formalin fixed paraffin embedded tissue section of the human kidney (NBP1-78269) using 1:100 dilution of ERG antibody. The signal was developed using HRP-DAB method which followed counterstaining of the cells with hematoxylin. The antibody generated mainly a nuclear staining with weak cytoplasmic positivity in the cells of the glomeruli and the tubular epithelial cells.
