Recognizes a protein of 185kDa, which is identified as c-erbB-2/HER-2/neu. Its epitope is localized in the extracellular domain. C-erbB-2/HER-2 is a member of the EGFR family. This monoclonal antibody is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. C-erbB-2/HER-2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary. |

Background
ErbB2/HER2 is one of the four members of the ErbB receptor family of transmembrane receptor-like tyrosine kinases (1). The kinase activity of ErbB2 can be activated without ligand if it is overexpressed, and by association with other ErbB proteins (2). Overexpression of ErbB2 is detected in almost 40% of human breast cancers (3). Binding of c-Cbl ubiquitin ligase to Tyr1112 of ErbB2 leads to poly-ubiquitination of ErbB2 and enhances its degradation (4). ErbB2 is one of the major targets for the treatment of breast cancer and other carcinomas.

- NBP2-34641AF647 and 488 is made to a recombinant fragment corresponding to the extracellular domain of the human HER2 protein. The epitope is localized in the extracellular domain.

I was curious what 100 tests corresponds to, concentration wise?
- 100 tests corresponds to 100ug as 1ug/test. The base product of this antibody (NBP2-34641) is supplied at 1mg/ml, so the vial will have 100ul of volume. The vial of conjugated antibody will contain at least 100ug of antibody.

I am curious whether the epitope of the antibody NBP2-34641AF647 is located intra- or extracellularly? I would like to use this antibody to sort cells and thus need to avoid any permeabilization or fixation steps.
- The immunogen used to generate this antibody is a recombinant extracellular domain of human HER-2. Unfortunately, I do not have access to the exact immunogen sequence as that information is proprietary.
