Application Notes
IHC: NBP1-02456 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. Use in Flow Cytometry was reported in a scientific publication (PMID: 23691119).

Background

DNA dependent Protein Kinase (also called DNAPK, DNPK1, HYRC1, Protein Kinase DNA Activated Catalytic Polypeptide, XRCC7 and p460) consists of the 460 kDa DNA PKcs and a heterodimeric regulatory complex comprised of p70 Ku and p80 Ku (Ku autoantigen). DNA PKcs is a nuclear protein serine/threonine kinase present in a wide variety of eukaryotic species. DNA PKcs phosphorylates transcription factors, Sp1, Oct-1, p53 and SV40 large T antigen. DNA PKcs is involved in repairing double stranded DNA breaks. At the onset of apoptosis, DNA PKcs is rapidly inactivated by cleavage of the catalytic subunit into smaller polypeptides. Proteolysis of DNA PKcs is inhibited by the cysteine protease inhibitors iodoacetamide and N-ethylmaleimide. Alternative splicing can occur for this protein to produce at least two isoforms.

Whole cell protein from Jurkat cells treated with and without 2 uM staurosporine as indicated was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 5 ug/ml anti-Caspase 3 in 1% milk, and detected with an anti-mouse HRP secondary antibody using a Femto sensitivity chemiluminescence reagent. Note the detection of both pro-caspase 3 at 35 kDa and the cleaved active caspase 3 at 15-17 kDa.

Western Blot: Chk2 Antibody (73C175.1.1) [NB100-56546] - Western Blot: Chk2 Antibody (73C175.1.1) - Azide Free [NBP2-27403] - analysis of Chk2 in 293 cell lysate. A protein band of approximate molecular weight of 60-62 kDa is detected with Chk2 antibody at 2 ug/ml. Image using the Azide Free form of this antibody.
Immunocytochemistry/Immunofluorescence: Chk2 Antibody (73C175.1.1) [NB100-56546] - Hek293 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. The cells were incubated with anti-Chk2 (73C175.1.1) at 5 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
