Western Blot: DIAPH1 Antibody [NB600-230] - Samples: Whole cell lysate (5, 15 and 50 ug for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-Diaphanous 1 antibody used for WB at 0.04 ug/ml (A) and 1 ug/ml (B) and used for IP at 3 ug/mg lysate. Diaphanous 1 was also immunoprecipitated by a prior lot of this antibody. Detection: Chemiluminescence with exposure times of 30 seconds (A) and 1 second (B).

Background

Diaphanous 1 is a member of the Formin homology (FH) family of cytoskeletal organizer proteins. It is required for cytokinesis and transcriptional activation of the serum response factor. It has also been suggested to contribute to localized actin polymerization and stress fiber formation, both by recruiting profiling to sites of Rho action and by cooperating with ROCK. The human Diaphanous 1 gene is associated with Premature Ovarian Failure (POF) and Nonsyndromic Deafness (DFNA1), an autosomal dominant, fully penetrant progressive hearing loss.
