Background
CD39 is an acidic glycoprotein which is a prototype member of the nucleoside 5'-triphosphate di-phosphohydrolase family. Initially it was identified as an activation marker for B lymphocytes and subsequently shown to be expressed on subsets of activated NK cells, T lymphocytes, epidermal langerhans dendritic cells and endothelial cells (Deaglio et al., 2011, Wu et al., 2006, Koziak et al., 2000). The gene for CD39 is mapped to chromosome 10q24 and consists of 6 isoforms (Gray et al., 1997). CD39 acts as an important plasma membrane-bound ecto-nucleotidase with pivotal roles in the hydrolysis of extracellular nucleoside di- and tri-phosphates by the endothelium and leucocytes. It also modulates vascular inflammation, cellular proliferation and migration and plays a crucial role in the regulation of the ADP-purinoreceptor P2Y1 [P2 (purinergic type-2)-receptor 1] function (Wang et al., 2012, Wu et al., 2006). The molecular weight of CD39 is approximately 78KD and comprises N- and C-terminal transmembrane-anchoring domains with short cytoplasmic tails, and a large glycosylated extracellular domain containing five ACR (apyrase conserved region) sequences. It also undergoes several post-translational modifications including proteolytical cleavage, N-glycosylation and constitutive palmitoylation. Palmitoylation greatly stabilizes membrane association, augments NTPDase (nucleoside triphosphate diphosphohydrolase) activity and targets the CD39 molecule, in association with P2Y1 receptors, to specialized membrane microdomains termed caveolae that are crucial in the regulation of signal transduction (Wu et al., 2006, Kittel et al., 1999). CD39 ectonucleotidase is the rate-limiting enzyme of a cascade leading to the generation of suppressive adenosine that alters CD4 and CD8 T cell and natural killer cell antitumor activities. Recent studies have supported CD39 as a promising therapeutic target in oncology and the development of CD39 antibody which could selectively inhibit Treg function without depletion (Bastid et al., 2013).
