Amyloid peptides, derived from amyloid precursor protein (APP), are thought to play a role in the development of the senile plaques associated with Alzheimer's Disease. The amyloid hypothesis presupposes that flaws in the processing of APP result in abnormally high levels of the longer, "stickier" forms of beta amyloid, known as ABeta42 and ABeta43, leading to aggregation of amyloid in the neuronal cell death and ultimately neuronal death. Mutations in the structure of ABeta40 and related peptides as well as in some of the enzymes involved in the processing of APP have been shown to alter the processing of APP. The sporadic (i.e., non-genetic) form of the disease, however, is far more common, caused by aging in concert with a number of both genetic and environmental risk factors.

  1. We have purchased beta amyloid protein (NBP1-44967), can you provide full details on the instructions for the reconstitution of this protein? We are going to use it in ELISA
    • During storage Abeta 1-42 will form insoluble aggregates. Abeta 1-42 is soluble in hexafluoroisopropanol (HFIP), DMSO, or bases such as 0.1% ammonia (aq), 50 mM TRIS - HCl, or 1mM NaOH in water, and under these conditions aggregated peptide will revert to the monomeric form. The Abeta 1-42 solutions in DMSO, or the above aqueous bases can then be diluted using various stocks solutions of appropriate buffers (10mM Tris, pH 7.4, and others). Peptide solubilized using HFIP should be evaporated, and if possible lyophilized once after reconstitution, as HFIP has been shown to have various effects on cells. Once properly treated with HFIP the 1-42 will be monomeric, soluble peptide that can then be reconstituted as above to induce fibrillation.
