Immunoprecipitation: BRCA1 Antibody (17F8) [NBP1-41185] - BRCA1 antibody 6B4 and BRCA1 antibody 17F8 was used for IP-WB assay. 6B4 alone (4 microgram), 17F8 alone (4 microgram), 6B4 plus 17F8 (2 microgram each), and mouse control normal IgG were used in an immunoprecipitation assay with MCF7 cell extract. Immunoprecipitated BRCA1 was detected in WB using BRCA1 antibody 6B4 at 1:1000 dilution. HeLa whole cell extract (20 microgram) was used as input in the Western blot assay.

<b>Genetic Strategies Validation. </b>Western Blot: BRCA1 Antibody (17F8) [NBP1-41185] - Wild-type (WT) and BRCA1 knockout (KO) HeLa cell extracts (30 ug) were separated by 5% SDS-PAGE, and the membrane was blotted with BRCA1 antibody [17F8] - ChIP grade diluted at 1:500. The HRP-conjugated anti-mouse IgG antibody was used to detect the primary antibody, and the signal was developed with Trident ECL plus-Enhanced.

<b>Independent Antibodies Validation. </b>Immunoprecipitation: Carm1 Antibody [NB200-342] - Detection of human CARM1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-CARM1 antibody NB200-342 (lot NB200-342-4) used for IP at 6 ug per reaction. CARM1 was also immunoprecipitated by a previous lot of this antibody (lot NB200-342-3) and another rabbit anti-CARM1 antibody. For blotting immunoprecipitated CARM1, NB200-342 was used at 0.04 ug/ml. Detection: Chemiluminescence with an exposure time of 3 seconds.

CARM1 is upregulated in HCC tissues and indicates poor clinical outcomes.A Western blotting analysis showed the expression of CARM1 in 35 pairs of HCC and adjacent normal liver tissues. Immunoprecipitation: p19ARF/CDKN2A Antibody (12-A1-1) [NB200-169] - Total cell lysates (250 ug) from NIH3T3 cells which have deleted the Arf gene (Lanes 5) and from NIH3T3 cells expressing an HA-tagged version of p19Arf (lanes 3, 4) were incubated with control rat IgG (Lanes 3), or NB200-169 (lanes 4, 5) and protein G- Sepharose. Precipitated proteins were resolved by SDS-PAGE and analysed by Western blotting using an anti-HA antibody. Blue box: in WT cells, XRCC1 protein complexes limit PARP1 engagement and activity during BER by promoting efficient hand-off of SSB intermediates to POL beta and LIG3, preventing PARP1 from impeding repair. Orange box: in XRCC1−/− cells, the absence of XRCC1 protein complexes results in excessive cycles of PARP1 association/activation at SSB intermediates, which impedes access by other BER enzymes and blocks their repair, resulting in SSB accumulation. If this scenario is sufficiently prolonged, such as at high levels of base damage, then this increased PARP1 engagement leads progressively to NAD+ depletion, declining PARP1 auto-ribosylation and dissociation, and accumulation of PARP1 in chromatin. PARP1 trapping in this scenario thus reflects both increased PARP1 association at SSB intermediates and subsequently decreased PARP1 dissociation, both of which impede BER in a manner reminiscent of chemical PARP inhibitors (pink box shown for comparison). Immunocytochemistry/Immunofluorescence: 53BP1 Antibody (1285C) [NBP2-54753] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with 53BP1 Antibody at 5 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) (Catalog #NB100-690) was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow (Intracellular): 53BP1 Antibody (1285C) [NBP2-54753] - An intracellular stain was performed on HeLa Cells with 53BP1 (1285C) antibody (Catalog #NBP2-54753) (blue) and a matched isotype control MAB1050 (orange). Cells were fixed with 4% paraformaldehyde, following fixation, cells were permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by rabbit IgG APC-conjugated secondary antibody (F0111, R&D Systems).

Immunocytochemistry/Immunofluorescence: 53BP1 Antibody (1285C) [NBP2-54753] - NIH3T3 cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-53BP1 Antibody [1285C] NBP2-54753 at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:1000 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: 53BP1 Antibody (1285C) [NBP2-54753] - An intracellular stain was performed on Ntera2 cells with 53BP1 Antibody [1285C] NBP2-54753 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1.0 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.
