TfR (Transferrin R) was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti- TfR (Transferrin R) Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 488 (Catalog # NB100-92243AF488) (green) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (dark blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/Immunofluorescence: CD31/PECAM-1 Antibody [NBP1-71663] - HUVEC cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with CD31/PECAM-1 Antibody at 5 ug/mL overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Immunohistochemistry Free-Floating: Calbindin D-28K Antibody [NBP2-50028] - Analysis of rat cerebellum section stained with chicken Calbindin D-28K pAb, dilution 1:2,000 (Green), and costained with rabbit MeCP2 pAb, dilution 1:5,000 (Red). Following transcardial perfusion with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. Calbindin D-28K, often used as a Purkinje cell marker, is prominently detected in dendrites and perikarya of these cells in the cerebellar molecular layer. The MeCP2 antibody selectively stains nuclei of neuronal cells.

Immunocytochemistry/ Immunofluorescence: Calbindin D-28K Antibody [NBP2-50028] - Myh14 localizes to apical membranes of Atp6v1b1‐positive intercalated cells in the distal tubule. Adult kidney sections were stained for calbindin‐D28K, Atp6v1b1, Aqp2, & Myh14 to identify intercalated cells within the distal & connecting tubule segments as well as to confirm Myh14 localization to the intercalated cell type. Cells with positive staining for Atp6v1b1 (red) can be found in tubule segments positive for calbindin‐D28K only (A–D) as well as in tubules positive for both calbindin‐D28K & Aqp2 (E–H); in both cases Atp6v1b1 staining is observed along the basolateral membrane indicating beta ‐ intercalated cell type (arrowhead, D&H). We also observed Atp6v1b1 staining in majority of the tubules along the apical membrane indicating the alpha ‐ intercalated cell type (arrowhead, I–L). Myh14 localized to the apical membrane of the intercalated cell type in the distal tubule (arrowhead, M–P). Immunocytochemistry/Immunofluorescence: KAT3B/p300 Antibody (RW105) [NB100-616] - HeLa cells were fixed in 4% paraformaldehyde for 10 minutes and permeabilized in 0.5% Triton X-100 in PBS for 5 minutes. The cells were incubated with anti-KAT3B/p300 Antibody (RW105) NB100-616 at 2 ug/ml overnight at 4C and detected with an anti-mouse Dylight 488 (Green) at a 1:1000 dilution for 60 minutes. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 100X objective and digitally deconvolved.

Flow Cytometry: KAT3B/p300 Antibody (RW105) [NB100-616] - An intracellular stain was performed on RAW 246.7 cells with KAT3B/p300 antibody (RW105) NB100-616PE (blue) and a matched isotype control NBP2-27287PE (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature. Cells were incubated in an antibody dilution of 1:200 for 20 minutes at room temperature. Both antibodies were conjugated to APC. A co-stain was performed using CD14 antibody (M5E2) NB100-77758PE.

Dual RNAscope ISH-IHC: CD14 Antibody (M5E2) [NB100-77758] - TNFA mRNA (red) and CD14 protein (green) were detected in formalin-fixed paraffin-embedded tissue sections of human malignant lymph node. ACD's Integrated Co-Detection Workflow was performed using ACD RNAScope Probe Hs-TNFA and CD14 antibody (M5E2) at 1:200 dilution. Tissue was stained on Leica Bond RX using RNAscope (TM) 2.5 LS Reagent Kit-RED, BOND Polymer Refine Detection (DAB) and Hematoxylin, BOND Polymer Refine Red Detection and Hematoxylin and RNAscope (TM) 2.5 LS Green Accessory Pack. Tissue was counterstained with 50% hematoxylin (blue).

Immunocytochemistry/Immunofluorescence: MCT1/SLC16A1 Antibody [NBP1-59656] - Monocarboxylic acid transporter 1 antibody was tested in HeLa cells with DyLight488 (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and Dylight 550 (red). General cytoplasmic and membrane staining was observed.

Western Blot: MCT1/SLC16A1 Antibody [NBP1-59656] - Monocarboxylic acid transporter 1 Antibody [NBP1-59656] - Jurkat cell lysate, concentration 2.5 ug/mL.

MCT1/SLC16A1 was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti- MCT1/SLC16A1 Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP1-59656AF647) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (dark blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunocytochemistry/ Immunofluorescence: MCT1/SLC16A1 Antibody - BSA Free [NBP1-59656] - Immunofluorescence detection of ion transporters & water transporters in differentiated & CST- SPs. Na+/K+-ATP isoform ATP1A1, water transporter Aquaporin 1, SLC4A11, Bicarbonate transporter NBCe1, Na+-H+ exchanger NHE1 & ZO-1 as an endothelial barrier to complements fluid transport, were all selectively expressed in differentiated SPs, but were almost null in the cell-state-transitioned SPs. Monocarboxylic acid transporter 4 (MCT4) was expressed in the former, but not in the latter SPs, whereas the isomer MCT1 was selectively expressed in the latter SPs, but not in the former SPs. Differentiated SPs were obtained from the same donor (two eyes, right & left mixed) & were incubated in the presence of 10-µM Y27632 (differentiated SPs, CD44-/ + proportion: 86.7%). CST-SPs were obtained from the separate donor (two eyes, right & left mixed) & were incubated in the presence of 10-µM Y27632 + 1-µM SB431542 (SB4) + 10-µM SB203580 (SB2) + 5-ng/mL epidermal growth factor (EGF). The differentiated SPs were fixed at 37 days of the second passage, & the CST SPs were fixed at 38 days of the second passage. Bars: 100 µm. Experiments were repeated four times.
