![Immunohistochemistry-Paraffin: Aquaporin-2 Antibody [NB110-74682] - Analysis of FFPE human kidney using 1:100 conc. of Aquaporin-2 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Lumenal staining of Aquaporin-2 in scattered tubules was observed. Frequencies of neutrophils, macrophages, & DCs in the colon were determined by flow cytometry. Cells were gated on CD45+CD3−CD4−CD11b+Ly6G+, CD45+CD3−CD4−CD11b+F4/80+, & CD45+CD3−CD4−CD11b+CD11c+ populations respectively. Representative flow cytometric figures were shown. The percentage of cells was presented as the mean ± SEM of four to six individual mice per group. *p < 0.05 in a one-way analysis of variance followed by Bonferroni correction. ![Immunocytochemistry/Immunofluorescence: RIPK3/RIP3 Antibody [NBP2-24588] - HeLa cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with RIPK3/RIP3 Antibody [NBP2-24588] at a 1:200 dilution overnight at 4C and detected with DyLight 488 (Green) at a 1:500 dilution. Alpha tubulin was used at a 1:1000 dilution as a co-stain and detected with DyLight 550 (Red) at a 1:500 dilution. Nuclei were detected with DAPI (Blue) at 2.0ug/ml in 1X PBS.
