Melphalan-sensitive MM cells (S) were treated with Vehicle or CASIN (5 μM) for 8 h. Cdc42 & Rac1 activities were measured using pull-down assay. (B) CASIN (5 μM) inhibits proliferation of both S & melphalan-resistant (LR5) MM cells. **P < 0.01 vs. S-Vehicle; ##P < 0.01 vs. LR5-Vehicle. (C) CASIN (5 μM) preferentially causes apoptosis of melphalan-resistant MM cells. S & LR5 cells were treated with or without CASIN or melphalan (Mel, 25 μM) or both for 2 days. The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) was performed using Aperio AT2 (Leica Biosystems). Punctate staining of neurons was observed.
