Flow Cytometry: Beclin 1 Antibody - BSA Free [NB500-249] - An intracellular stain was performed on U-87MG cells with Beclin 1 Antibody NB500-249 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Beclin 1 was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti-Beclin 1 Affinity Purified Polyclonal Antibody conjugated to FITC (Catalog # NB500-249F) (green) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (dark blue). Cells were imaged using a 100X objective and digitally deconvolved.

PBMC were isolated from whole blood and stained for surface markers before cells were fixed, permeabilised and stained for TLR-9. FSC and SSC were first used to gate out debris and SSC-A and SSC-H was utilized to eliminate duplicates. Further gating was done on CD45 and CD19, to target B cells. To separate between the different B cell populations, we gated on CD27 and IgD (D), followed by TLR-9 expression on these subsets. Data from one representative patient is shown. THP-1 human acute monocytic leukemia cell line was stained with Mouse anti-TLR9 (26C593.2) Protein-G purified Monoclonal Antibody conjugated to Janelia Fluor® 549 (Catalog # NBP2-24729JF549, blue histogram) or matched control antibody (orange histogram).

Alveolar epithelial cells (AECs), fibroblasts and fibrocytes all express Toll-like receptor-9. AECs, fibroblasts, fibrocytes and whole lungs were isolated from Balb/c, mice and total RNA prepared and analyzed for TLR-9 expression. The data were normalized to the expression level of TLR-9 in dendritic cells, which was set to 1 (n = 3/group). No TLR-9 message was amplified in cells from TLR-9-/- mice. (B, C) Immunohistochemistry was performed on paraffin wax-embedded whole-lung tissue from normal Balb/c mice. (B) Control staining with secondary antibody only; (C) diffuse TLR-9 staining, with notable staining in the bronchial epithelium, at the alveolar junctions and the interstitium. Fibroblasts (E) and AECs (G) were isolated from Balb/c mice and fixed. Immunofluorescence staining for TLR-9 shows that both cell types expressed TLR-9. Immunohistochemistry: TLR7 Antibody [NBP2-24906] - Expression of the TLRs 5 and 7 were evaluated in the PDAC tissue by their staining intensity using TLR5 antibody (NBP2-24787) and TLR7 antibody (NBP2-24906). Note negative staining in E and G; positive staining in F and H. For TLR7 immunopositivity was detectable in the cytoplasm with no notable membranous or nuclear positivity. TLR5 showed distinctive nuclear positivity, and detectable positivity in the cytoplasm in some samples. Immunohistochemistry-Paraffin: Beclin 1 Antibody [NB110-87318] - Analysis of a FFPE tissue section of human breast cancer using Beclin 1 antibody (Lot 71917) at 1:300 dilution. The primary antibody bound to Beclin 1 antigen in the tissue section was detected using a HRP labeled secondary antibody and DAB reagent. Nuclei of the cells were counterstained with hematoxylin. Cytoplasmic staining can be seen in the cancer as well as the stromal cells. The signal was strongest in a subset of stromal cells which appears to be the cancer associated fibroblasts.

Western Blot: Beclin 1 Antibody [NB110-87318] - Basal levels of autophagy in the hypothalamus of BACHD mice. The expression levels of selected autophagy markers were analyzed by Western blot in hypothalamic tissue from BACHD mice at 2, 6 and 12 months of age and compared with gender- and age-matched controls (n=3-6/group). The data showed a substantial maintenance of the autophagy flux, despite the presence of a metabolic phenotype in the BACHD mice already at early time points. Analysis of immune cells infiltration in cerebellum was performed at 4 weeks by immunohistochemistry using antibodies against (A) CD4, a marker of T lymphocytes (F(3,8) = 4.511, p < 0.01) and (B) IBA1, a marker of meningeal and perivascular macrophages (K-W(4,10) = 27.76, p < 0.0001). (C) Double immunofluorescence with anti-CD4 and anti-CX3CR1 as markers of autoreactive CD4+CD28− T lymphocytes was performed and quantified (K-W(4,5) = 8.711, p < 0.05). Immunohistochemistry: TLR7 Antibody (4G6) [NBP2-27332] - Analysis of a FFPE tissue section of human brain using 1:200 dilution of TLR7 [4G6] antibody. miR-298-5p and miR-100-5p enter microglia, co-localize to their endosomes, and bind directly to human TLR8. a C57BL/6 microglia were incubated with 40 μg/ml pHrodo Red Dextran serving as endosomal marker for 20 min. Western Blot: TLR3 Antibody [NB100-56571] - A TLR3 agonist polyinosinic‐polycytidylic acid (poly(I:C)) induced autophagy in cultured cardiomyocytes through a TRIF‐dependent pathway. (A) Poly(I:C) increased autophagy markers in cultured H9c2 rat ventricular cells. (B) Poly(I:C) stimulated autophagosome formation but did not affect autophagic flux. Primary cultured neonatal rat ventricular myocytes (NRVMs) were transfected with a tandem mRFP‐GFP‐LC3 adenovirus for 24 hrs, followed by treatment with poly(I:C) (100 μg/ml, 4 hrs). Autophagosomes & autolysosomes were, respectively, visualized as yellow‐ & red‐only punctas under a confocal microscope. (C) An autophagic flux inhibitor chloroquine (CQ) induced accumulations of LC3‐II & p62/SQSTM1 proteins in H9c2 myocytes receiving poly(I:C) (100 μg/ml, 4 hrs). CQ was applied at 10 μM, immediately prior to poly(I:C). (D) Effects of indicated siRNA on poly(I:C)‐induced changes in autophagy markers in NRVMs. All the siRNAs were transfected at 50 nM for 48 hrs, & poly(I:C) was added at 100 μg/ml for 4 hrs before cell harvest.
