U266 cells were treated with 100 uM of control (A & B) or NBD peptide (C & D) for 12 hr, cytospun, plated on glass slides, air dried for 1 hr at room temperature and fixed with cold acetone. Slides were blocked with 5% normal goat serum for 1 hr and then incubated with rabbit polyclonal anti-human p65 antibody (A & C) followed by Ig-Alexa 594 second step. ![IKK gamma Inhibitor Peptide Set [NBP2-26504] - The IKK complex consists of IKKalpha + IKKbeta+ NEMO. NEMO (IKKgamma/NF-kB Essential MOdulator) is a scaffold protein required for the kinase activity of IKKalpha + IKKbeta. NBD (NEMO Binding Domain) inhibits formation of the IKK complex (IKKalpha + IKKbeta+ NEMO) by binding to NEMO and blocking IKK complex formation. In this regard, NBD is considered to be an 'IKKalpha and IKKBeta decoy' Blocking IKK complex formation suppresses downstream events in the NF-kB signaling pathway that rely on the IKK complex including phosphorylation of IkBalpha, and subsequence IkBalpha ubiquination, degradation and release of NF-kB. VMN neurons were identified in situ for glutamate decarboxylase65/67 (GAD65/67)- (top row, Panel 1A) or neuronal nitric oxide (nNOS)-immunoreactivity (-ir) (bottom row, Panel 2A); representative GAD65/67- or nNOS-ir-positive neurons are indicated by blue arrows. Areas shown in Panels 1A & 2A were rephotographed after positioning of a continuous laser track (depicted in green) around a single GAD65/67-ir (Panel 1B, blue arrow) or nNOS-ir neuron (Panel 2B, blue arrow) & subsequent ejection of the encircled cell by laser pulse (Panels 1C & 2C). Note that this microdissection technique causes negligible destruction of surrounding tissue & minimal inclusion of adjacent tissue. (A,B) Exposure of microglia to cART resulted in a significant decrease in expression of lysosomal-associated membrane protein 2 (LAMP2) at 6 to 24 h post-treatment. (C) Representative bar graph showing cART-mediated significantly increased lysosomal membrane permeabilization (LMP) (24 h).
