Western Blot: WHSC1 Antibody (PSH02-39) [NBP3-32901] - Western blot analysis of WHSC1 on different lysates with Rabbit anti-WHSC1 antibody (NBP3-32901) at 1/1,000 dilution. Lane 1: HeLa cell lysate Lane 2: HEK-293 cell lysate Lane 3: Neuro-2a cell lysate Lane 4: NIH/3T3 cell lysate Lane 5: PC-12 cell lysate Lysates/proteins at 20 ug/Lane. Predicted band size: 66 kDa Observed band size: 66 kDa Exposure time: 1 minute; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32901) at 1/1,000 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: WHSC1 Antibody (PSH02-39) [NBP3-32901] - Immunohistochemical analysis of paraffin-embedded Rat testis tissue with Rabbit anti-WHSC1 antibody (NBP3-32901) at 1/200 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32901) at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

WHSC1 encodes a protein that contains four domains present in other developmental proteins: a PWWP domain, an HMG box, a SET domain, and a PHD-type zinc finger. It is expressed ubiquitously in early development. Wolf-Hirschhorn syndrome (WHS) is a malformation syndrome associated with a hemizygous deletion of the distal short arm of chromosome 4. This gene maps to the 165 kb WHS critical region and has also been involved in the chromosomal translocation t(4;14)(p16.3;q32.3) in multiple myelomas. Alternative splicing of this gene results in multiple transcript variants encoding different isoforms. Some transcript variants are nonsense-mediated mRNA (NMD) decay candidates, hence not represented as reference sequences. [provided by RefSeq]
