• Application Notes: WB: Preliminary experiments gave bands at approx. 50 kDa band and 26 kDa band in lysates of HEK293, A549 and HeLa after 0.5 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 93.5 kDa band according to NP_006438.1. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the bands).

Background

Ubiquitin (Ub) is a highly conserved protein found ubiquitously in eukaryotic organisms. The conjugation of ubiquitin to proteins is an important means to regulate protein activity for many cellular processes by tagging them for degradation. Removal of Ub can rescue proteins from degradation. This is accomplished by the ubiquitin-specific processing protease (UBP) family of enzymes. Ubiquitin specific processing protease 16 (USP16) is a member of this family. USP16 activity is cell cycle regulated via phosphorylation and participates in cell cycle control by influencing mitotic chromosome condensation and histone 2A (H2A) deubiquitination.
