Application Notes
NB 300-104 can be used in Western blot where a doublet is seen at ~ 80 kDa representing synapsin Ia and Ib. Immunofluoresence, Immunoprecipitation (1 ug will quantitatively immunoprecipitate all synapsin in 200 ug of rat brain.) May work in IHC-Paraffin but this has not been tested. Differential distribution of the synapsins in the rat olfactory bulb. Has been reported in the literature to work in WB (rat cortex), IHC (cryosections) on rat olfacotry bulb. (PMID: 8283238) Use in Immunohistochemistry free floating reported in multiple pieces of scientific literature.

Background

Synapsin I, also called Synapsin-1 or SYN1, belongs to Synapsin family (Synapsin I, Synapsin II, Synapsin III) of evolutionarily conserved phospho-proteins which are associated with the cytosolic side of the synaptic vesicles and tether the latter to actin cytoskeleton, thus forming a reserve pool. Synapsins represents the most abundant of neuron-specific phosphor-proteins, consisting 9% of the total amount of all vesicle proteins in neurons wherein they play a critical role in the regulation of neurotransmitter release. Synapsin I is present in axon terminals and plays a role in regulating the secretion of neurotransmitters; thus, it influences synaptic plasticity by regulating pre- and post-synaptic vesicular release. Synapsins I and II are typically found in mature synapses, while synapsin III is often attributed to developing synapses with lesser overall expression. Phosphorylation at Ser-9 residue results in dissociation of Synapsins I from synaptic vesicles which is important for synaptic vesicle neurotransmitter release. Mutations in Synapsin I have been linked to Epilepsy X-linked, with variable learning disabilities and behavior disorders (XELBD) which is a neurologic disorder characterized by variable combinations of epilepsy, learning difficulties, macrocephaly, and aggressive behavior.

  1. How many days do you recommend culturing primary neurons to get a good Synapsin I signal? Can you please send the protocol used to validate this antibody in immunofluorescence?
    • It was validated in IF by a collaborator, so I do not have a specific protocol to offer. However I can tell you that the neurons were cultured for 14 days.
