Western Blot: SUFU Antibody - Azide and BSA Free [NBP2-61114] - Detection of Sufu, phosphorylated S352 Sufu and HA after immunoprecipitation of HA tagged Sufu Wild Type and Sufu S352A from HEK293T cells. This shows that the anti-phospho-sufu antibody is specific and recognises only the phosphorylated form of Sufu but not a mutant where S352 is mutated to alanine. Also a second antibody recognises Sufu after immunoprecipitation independent of the phosphorylation status.

Immunofluorescence: Suppressor of Fused Antibody - Azide and BSA Free [NBP2-61114] - Immunofluorescence staining of the indicated proteins in cyliated mouse embryonic fibroblasts (MEFs) and MEFs knockout for Sufu-/-. The images show pSufu accumulates at the base of the cilium, similarly to the phosphorylated S342 described before. In this case, MEFs lacking Sufu do not show the same signal indicating specificity.

Reactivity Notes

Anti-phospho-Sufu polyclonal antibody recognises wild type sufu which is phosphorylated at serine 352. The antibody specificity for the phosphorylated version has been confirmed; it does not react against a non-phosphorylated version of Sufu which has serine 352 substituted by alanine.

Background

The Hedgehog (Hh) signaling pathway has conserved roles in development of species ranging from Drosophila to humans. Human supressor of fused HSu(Fu) acts as a negative regulator in the hedgehog signaling (SHH) pathway by down-regulating GLI1-mediated transactivation of target genes (1). HSu(fu) was found to repress activity of the zinc-finger transcription factor Gli, which mediates Hedgehog signaling in vertebrates, and to physically interact with Gli, Gli2 and Gli3 (2). The sonic hedgehog signaling pathway directs the embryonic development of diverse organisms and is disrupted in a variety of malignancies. Several of mutations encode truncated proteins that are unable to export the GLI transcription factor from nucleus to cytoplasm, resulting in the activation of SHH signaling. SUFU is a newly identified tumor-suppressor gene that predisposes individuals to medulloblastoma by modulating the SHH signaling pathway through a newly identified mechanism (3).
