SIRT6 Antibody (JJ20-69)

Western Blot: SIRT6 Antibody (JJ20-69) [NBP2-67854] - Analysis of SIRT6 on Hela cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: SIRT6 Antibody (JJ20-69) [NBP2-67854] - Staining SIRT6 in NIH/3T3 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: SIRT6 Antibody (JJ20-69) [NBP2-67854] - Analysis of paraffin-embedded human colon carcinoma tissue using anti-SIRT6 antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 20 minutes. The tissues were blocked in 1% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (1/400) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: SIRT6 Antibody (JJ20-69) [NBP2-67854] - Analysis of Hela cells with SIRT6 antibody at 1/50 dilution (red) compared with an unlabelled control (cells without incubation with primary antibody; black). Alexa Fluor 488-conjugated goat anti rabbit IgG was used as the secondary antibody

Background

Sirtuins regulate epigenetic gene silencing. SIRT6 (Mono-ADP-ribosyltransferase sirtuin-6)--a mouse Sir2 homolog--is a chromatin-associated nuclear ADP-ribosyltransferase protein. It appears to be involved in DNA repair and may also play a role in the ageing process.
