Plectin Antibody (SY29-04)

Knockout Validated: Plectin Antibody (SY29-04) [NBP2-67452] - Western blot using lysates from U20S parental cell line and Plectin-1 knockout U20S cell line (KO), collected in RIPA buffer. Nitrocellulose membrane was probed with Mouse Anti-Human/Mouse Plectin Monoclonal Antibody (Catalog # NBP2-67452) at a 1:1000 dilution O/N at 4C, followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody and ECL detection. A specific band was detected for Plectin-1 (as indicated) in the parental U20S cell line, but is not detectable in knockout U20S cell line. Knockout Validated: Plectin Antibody (SY29-04) [NBP2-67452] - Parental and KO cells were transfected with a GFP or mCherry plasmid, respectively. Both cell lines were mixed and plated to a 1:1 ratio on coverslips. Cells were stained with an anti-Plectin antibody and with the corresponding Alexa-fluor 647 coupled secondary antibody. Acquisition of the green (parental), red (KO) and far-red (antibody staining) channels was performed, and representative images are shown. Background

Plectin, a 500-kDa intermediate filament binding protein, is localized to the cytoplasm and acts to both stabilize and give strength to cells via cross-linking (1). Plectin cross-links intermediate filaments to microtubules and microfilaments, and anchors intermediate filaments to the plasma and nuclear membranes (2,3). Plectin binds many proteins, including actin, vimentin, and Lamin B. Phosphorylation by PKA or PKC results in decreased binding to Lamin B, and phosphorylation by PKA enhances the plectin-vimentin interactions (4).
