Application Notes
Predicted molecular weight 30 kDa. Western Blot: incubation 60 min Sample preparation: Resuspend approx. 50 mil. cells in 1 ml cold Lysis buffer (1% laurylmaltoside in 20 mM Tris/Cl, 100 mM NaCl pH 8.2, 50 mM NaF including Protease inhibitor Cocktail). Incubate 60 min on ice. Centrifuge to remove cell debris. Mix lysate with non-reducing SDS-PAGE sample buffer. Non-Reducing conditions. 12% separating SDS-PAGE gel.

Background

The adaptor protein NTAL (Non-T cell Activation Linker; human 30 kDa, mouse 25 kDa), alternatively termed as linker for activation of B cells (LAB), is expressed in plasma membrane lipid rafts of B cells, mast cells, monocytes and NK cells. The engagement and activation of antigen receptors on B and T lymphocytes initiates signal transducing pathways. These pathways are vital for the successful initiation of an adaptive immune response and it is following the cross-linking of the B cell receptor (BCR) or high-affinity FE receptors of myeloid cells, that the tyrosine phosphorylation of NTAL occurs, enabling the formation of complexes between NTAL and further signaling molecules Grb2, Gab1, Sos1 and c-Cbl. Antigen-initiated activation events include tyrosine phosphorylation of two transmembrane adaptor proteins, LAT and NTAL; however NTAL is functionally and topographically different from LAT. Recent studies have indicated, that the evidence for functional homology between LAT and NTAL/LAB, and the possibility that the differences between them might lead to specific clinical therapeutics to manipulate immune cell responses.
