Background

The inner mitochondrial membrane forms internal compartments known as cristae and holds the essential proteins (NADH dehydrogenase, cytochrome c reductase, cytochrome c oxidase) involved in the respiratory chain, facilitating the production of ATP from NADH.

Comments

CommentsPer the Novus Innovator's Reward, we decided to test whether this Mitofilin Supernatant product would work well for Western Blot, seeing that it worked well for other applications in our target species Rat.
The gel was loaded with 15 ug per lane of brain lysate. PVDF membranes were blocked with 5% milk for 1 h, and incubated with Mitofilin H6/C12 overnight at 4C. Anti mouse-HRP secondary was applied for 2h at room temp, with blots developed using high-sensitivity Supersignal West Femto ECL reagent. We attempted using this antibody at 1:200 and 1:40 (shown here), with no signal detected. The same blot was successfully tested with known good antibody, which worked, pointing to successful transfer and detection.

Mitofilin and the Mitochondrial Inner Membrane Organizing System (MINOS)
Mitofilin was originally described as a heart muscle protein due to its high expression in the heart. Recently, analysis of the human heart mitochondrial proteome demonstrated that Mitofilin is one of the most abundant mitochondrial proteins (1). Rese...  Read full blog post.
