Recognizes a single protein of 40kDa, identified as the CD305 (LAIR1). It is an inhibitory receptor, expressed on the majority of peripheral mononuclear cells, including natural killer (NK) cells, T-cells, B-cells, monocytes, and dendritic cells. Highly expressed in naive T-cells and B-cells but no expression on germinal center B-cells. This MAb is expressed on CD34 positive hematopoietic progenitors. Inhibitory receptors regulate the immune response to prevent lysis of cells recognized as self. The gene is a member of both the immunoglobulin superfamily and the leukocyte-associated inhibitory receptor family. LAIR1 has been identified as an anchor for tyrosine phosphatase SHP-1, and may induce cell death in myeloid leukemias. Alternative splicing results in multiple transcript variants. This MAb is identical to LAIR-1, based on the fact that it not only precipitated a protein of 40kDa but it also binds specifically to LAIR-1 cDNA transfected COS7 cells as well as recognizes LAIR-1 fusion protein in ELISA. LAIR-1 expresses on hematopoietic progenitor, implicating its role in the regulation of hematopoiesis at early stage. |

Background
LAIR1 is a transmembrane glycoprotein(approx. 32kDa) expressed on the majority of human peripheral blood mononuclear leukocytes including T cells, B cells, NK cells, macrophages and dendritic cells. LAIR1 recruits SHP1 and SHP2 tyrosine phosphatases upon activation, and cross-linking of the LAIR1 molecule on NK cells results in strong inhibition of NK cell mediated cytotoxicity. This clone is reported to inhibit NK cell triggering via CD16 molecules.
