Human JunB/AP-1 Luciferase - (LUCPorter™) Stable Reporter Cell Line Summary
Description
The AP-1 LUCPorter™ Reporter Cell Line can be used for studying the AP-1 mediated signaling pathways as well as screening of agonists/antagonists/signaling inhibitors related with AP-1 dependent cell activation.
The AP-1 transcription factors are homo- or hetero-dimers that consist of proteins belonging to a group of structurally and functionally related members of the Jun family (c-Jun, JunB and JunD), the Fos family (c-Fos, FosB, Fra-1 and Fra-2) and the subfamilies of ATF (ATFa, ATF-2 and ATF-3) and JDP (JDP-1 and JDP-2). AP-1 can be activated by a variety of stimuli including cytokines, growth factors, stress and microbial infections, subsequently modulating various cellular processes including differentiation, cell cycle progression and apoptosis. Functional activity of the reporter cell line has been validated by induction of the AP-1-responsive luciferase activity with different stimuli, in which the cell line is specifically activated by phorbol 12-myristate 13-acetate (PMA) (Figure 1). Dose dependent activation of the AP-1-response element in the cell line has also been tested with various PMA doses (Figure 2).

Immunogen
The AP-1 LUCPorter™ Reporter Cell Line is a stably transfected cell line which expresses an optimized Renilla luciferase reporter gene under the transcriptional control of the activator protein 1 (AP-1).
Assume all cultures are hazardous since they may harbor latent viruses or other organisms that are uncharacterized. The following safety precautions should be observed. - Use pipette aids to prevent ingestion and keep aerosols down to a minimum. - No eating, drinking or smoking while handling the AP-1 reporter cell line. - Wash hands after handling the AP-1 reporter cell line and before leaving the lab. - Decontaminate work surface with disinfectant or 70% ethanol before and after working with the cell line. - All waste should be considered hazardous. - Dispose of all liquid waste after each experiment and treat with bleach.

Background
The c-Jun proto-oncogene was first identified as the cellular homolog of the avian sarcoma virus v-Jun oncogene. The c-Jun protein along with c-Fos is a component of the AP-1 transcriptional complex. c-Jun can form either Jun/Jun homodimers or Jun/Fos heterodimers via the leucine repeats in both proteins. Homo- and heterodimers bind to the TGACTCA consensus sequence present in numerous promoters and initially identified as the phorbol ester tumor promoter response element (TRE). Two additional genes, Jun B and Jun D have been shown to be almost identical to c-Jun in their C-terminal regions, which are involved in dimerization and DNA binding, whereas their N-terminal domains, which are involved in transcriptional activation, diverge. All three form heterodimers among themselves and with c-Fos and other members of the Fos gene family.
